目的 观测表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对人大肠癌SW620细胞增殖及SW620细胞表达PAK1的影响.方法 用终浓度为40 μmol/L、60 μmol/L和80 ixmo1/L的EGCG干预人大肠癌细胞株SW620,在0、24、48和72 h不同时间点上用噻唑蓝(MTT)比色法检测EGCG对SW620细胞生长的影响;并以SW620细胞空白组为对照,用蛋白免疫印迹(Westerm blot)检测不同EGCG作用48 h时SW620细胞PAK1的表达水平变化.结果 EGCG对大肠癌SW620细胞增殖有显著的抑制作用,而且与SW620细胞空白对照组相比.EGCG处理组细胞的PAK1蛋白表达受到显著抑制.结论 EGCG抑制大肠癌SW620细胞增殖的机制可能与EGCG通过调控大肠癌细胞的PAK1表达相关.
目的 觀測錶沒食子兒茶素沒食子痠酯(epigallocatechin-3-gallate,EGCG)對人大腸癌SW620細胞增殖及SW620細胞錶達PAK1的影響.方法 用終濃度為40 μmol/L、60 μmol/L和80 ixmo1/L的EGCG榦預人大腸癌細胞株SW620,在0、24、48和72 h不同時間點上用噻唑藍(MTT)比色法檢測EGCG對SW620細胞生長的影響;併以SW620細胞空白組為對照,用蛋白免疫印跡(Westerm blot)檢測不同EGCG作用48 h時SW620細胞PAK1的錶達水平變化.結果 EGCG對大腸癌SW620細胞增殖有顯著的抑製作用,而且與SW620細胞空白對照組相比.EGCG處理組細胞的PAK1蛋白錶達受到顯著抑製.結論 EGCG抑製大腸癌SW620細胞增殖的機製可能與EGCG通過調控大腸癌細胞的PAK1錶達相關.

Objective To investigate the effects of epigallocatechin-3-gallate (EGCG) on the proliferation of SW620 cells and the expression of PAK1 gene. Methods Human colonic cancer cell line SW620 was treated with EGCG at 40, 60 and 80 μmol/L and cultured in RPMI 1640 medium for 0, 24, 48 and 72 h. The proliferation of SW620 cells was observed by MTT assay before and after EGCG treatment, and the expression of PAK1 protein was observed by Western blotting. Results SW620 cells treated with EGCG displayed a slowed growth in comparison with the control cells, and the growth rate decreased with the increase of EGCG concentration. PAK1 protein expression was lowered in SW62Q, cells after EGCG treatment for 48 h. Conclusion EGCG can inhibit the proliferation and partially reduce the expression of PAK1 protein in SW620 cells.
