目的 探讨乳香酯对氧化型低密度脂蛋白(ox-LDL)所致的巨噬细胞单核细胞趋化因子(MCP-1)释放的影响及相关分子机制.方法 以ox-LDL、ox-LDL加不同浓度的乳香酯处理人单核细胞THP-1,采用酶联免疫吸附测定细胞上清液中MCP-1的浓度,免疫印迹法检测细胞质中p65/NF-κB和IκBα的表达水平以及细胞核内p65/NF-κB的表达水平.结果 乳香酯能够显著减少ox-LDL所致的巨噬细胞MCP-1的表达,且p65/NF-κB的核移位减少,IκBα的降解水平明显降低.结论 乳香酯减少ox-LDL所致的巨噬细胞MCP-1的表达与其调节磷酸化IκBα的表达和NF-κB的核-浆转位有关.

목적 탐토유향지대양화형저밀도지단백(ox-LDL)소치적거서세포단핵세포추화인자(MCP-1)석방적영향급상관분자궤제.방법 이ox-LDL、ox-LDL가불동농도적유향지처리인단핵세포THP-1,채용매련면역흡부측정세포상청액중MCP-1적농도,면역인적법검측세포질중p65/NF-κB화IκBα적표체수평이급세포핵내p65/NF-κB적표체수평.결과 유향지능구현저감소ox-LDL소치적거서세포MCP-1적표체,차p65/NF-κB적핵이위감소,IκBα적강해수평명현강저.결론 유향지감소ox-LDL소치적거서세포MCP-1적표체여기조절린산화IκBα적표체화NF-κB적핵-장전위유관.
Objective To explore the effect of gum mastic on the MCP-1 production in macrophages stimulated by oxidized low-density lipoprotein(ox-LDL).Methods THP-1 macrophage were treated with ox-LDL alone or accompanied with different concentrations of gum mastic,the level of MCP-1 in cell culture supernatants was measured by ELISA,the level of nuclear pool and plasma of p65/NF-κB protein was examined by Western blot for NF-κB nuclear translocation analysis,and the degradation of IκBα was detemined by western blot.Results Gum mastic inhibited the MCP-1 release induced by ox-LDL,and gum mastic also inhibited NF-κB translocation and the IκBα degradation.Conclusions The effect of gum mastic block the production of the MCP-1 is related to inhibiting the NF-κB pathway.
