建立了细菌外排泵抑制剂的筛选与活性跟踪方法.准备2个平板,一个为普通营养琼脂平板,另一个为舍小蘖碱的普通营养琼脂平板,通过比较两个平板含药纸片周围抑菌圈的直径大小判断筛选结果,方法可靠稳定.筛选发现某霉菌提取物对细菌外排泵有抑制活性,经活性跟踪分离,得到单体化合物,经NMR鉴定为4′,5,7-三羟基异黄酮.方法简便易行,成本低,适宜于对大批样本进行快速筛选并在分离时进行活性跟踪.

To establish the method of screening for and bioassay-guided isolation of bacterial efflux pump inhibitors.Two plates were prepared.One was nutrient agar plate and another was also nutrient agar plate but contained berberine.The determination of the screening results was made by the measurements of the diameters of the inhibition zone around the discs that to be pasted on the surface of the two phtes and contained test samples.The method was reliable and steady.The extract of one kind of the mould showed the activity to inhibit bacterial efflux pump.A monomeric compound was purified by bioassay-guided isolation and identified as 4′,5,7-trihydroxyisoflavone by 1H and 13C of NMR.The screening method for bacterial efflux pump inhibitors was very simple and cheap for practice.It is suitable for who wanted to screen for bacterial efflux pump inhibitors from large quantities of samples.Also it is a good bioassay in isolation.
