目的:建立反相高效液相色谱法测定清热解毒方中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚含量的方法.方法:采用HPLC法,色谱柱:Kromasil C18(250 mm×4.6 mm,5μm),以甲醇-0.2%磷酸(85:15)为流动相,流速:1.0 mL·min-1,检测波长:254 nm,柱温:35℃.结果:芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚分别在1.0125~16.2μg、1.025~16.4μg、1.025~16.4μg、1.025~16.4μg、0.625~10 μg范围内呈良好线性关系,相关系数分别为0.999 1,0.999 8,0.999 7,0.999 4,0.999 6,平均回收率分别为101.4%,102.0%,101.7%,102.1%,100.8%.结论:该方法操作简单、准确,且重复性好,可作为清热解毒方质量的控制指标之一.

Objective: To develop an HPLC method for determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in QING RE JIE DU entract. Methods: The analysis was carried out on a Kromasil C18 (250 mm×4.6 mm,5 μm)column eluted with a mobile phase of methanal-water containing 0.2% phosphoric acid solution(V : V=85 : 15). The flow rate was 1.0 mL· min-1 and column temperature was 35 ℃. The detection wavelength was set at 254 nm.Results: The calibration curve was linear within the range of 1.012 5~16.2,1.025~16.4,1.025~16.4,1.025~16.4 and 0.625~10 μg for aloe-emodin, rhein, emodin, chrysophanol and physcion, respectively. The average recoveries were 101.5% ,99.2%, 101.9%, 101.5%, 101.4% for the abore five conponents, respectively. Conclusion: This method is convenient, accurate and reproducible for determination of aloe-emodin, rhein, emodin, chrysophanol and phascion in QING RE JIE DU extraet.
