目的 探讨环氧化酶-2(COX-2)、骨桥蛋白(OPN)在人脑膜瘤中的表达及临床意义.方法 选择脑膜瘤患者40例作为观察组,10例健康体检者作为对照组.采用ELISA法测定COX-2和OPN的浓度;采用免疫组化技术检测术后40例脑膜瘤组织及10例正常脑膜组织,COX-2、OPN及基质金属蛋白酶-9(MMP-9)的表达.结果 侵袭组中COX-2、OPN的血清含量高于非侵袭组,差异有统计学意义(P<0.01);同样COX-2、OPN及MMP-9蛋白在侵袭组脑膜瘤中的表达均高于非侵袭组(P<0.01).在侵袭性脑膜瘤中COX-2、OPN和MMP-9两两比较,其表达均具有显著相关性.结论 COX-2、OPN与脑膜瘤的侵袭性密切相关,一定程度上可作为评价脑膜瘤侵袭性及预后的辅助指标,通过术前血清学检测、术后免疫组化检查可判断脑膜瘤的潜在侵袭性,指导临床治疗方案的制定,提高疗效水平.

Objective To investigate the expression of cyclooxygenase-2 (COX-2) and osteopontin( OPN ) in meningioma and the clinical significance.Methods Forty cases of meningioma were choosed as observation group,and 10 cases of health people were choosed as control group.The serum concentrations of COX-2 and OPN were detected by enzyme linked immunosorbent assay.The expression levels of OPN,COX-2 and MMP-9 were detected by immunohistochemical S-P method in 40 cases of meningiomas and 10 cases normal brain tissue.Results The serum concertrations of OPN and COX-2 in invasive meningioma were higher than those in non-invasive ( P < O.O1 ).The expression levels of COX2,OPN and MMP-9 in invasive meningioma was higher than that in non-invasive meningioma (P <0.01 ).There were significant relationps between COX-2,OPN and MMP-9 in invasive meningioma.Conclusions COX-2,OPN are closely correlated with meningioma invasiveness,indicating that COX-2,OPN can evaluate invasiveness and prognostic of meningioma.Through detecting of COX-2,OPN in the samples of meningioma,we can judge the potential invasiveness of meningioma,and institute a right treatment plan to improve the curative effect of tumor patients.
