目的 探讨中性粒细胞相关载脂蛋白(NGAL)在乳腺癌的表达及其临床意义.方法 乳腺癌手术切除标本47例,应用逆转录-聚合酶链反应(RT-PCR)检测NGAL mRNA相对表达量;应用免疫组织化学方法,分析NGAL蛋白表达情况.结果 乳腺癌组织中NGAL在乳腺癌组织中NGAL mRNA的表达水平为0.42±0.19,显著高于癌旁组织(P<0.01);NGAL蛋白在癌组织大多表达为~卅水平,癌旁正常组织则大多表达在-~+水平,NGAL蛋白在癌组织表达显著高于癌旁正常组织(P<0.01).NGAL mRNA和蛋白表达与淋巴结转移、病理类型、ER、HER-2状态明显相关(P<0.05),而与患者年龄、肿瘤大小、组织学分级、TNM分期、PR状态无明显相关(P>0.05).结论 NGAL的表达与乳腺癌淋巴结转移和内分泌治疗耐药等相关,NGAL可能是乳腺癌早期诊断、监测复发和判断预后的潜在生物学参考指标.

목적 탐토중성립세포상관재지단백(NGAL)재유선암적표체급기림상의의.방법 유선암수술절제표본47례,응용역전록-취합매련반응(RT-PCR)검측NGAL mRNA상대표체량;응용면역조직화학방법,분석NGAL단백표체정황.결과 유선암조직중NGAL재유선암조직중NGAL mRNA적표체수평위0.42±0.19,현저고우암방조직(P<0.01);NGAL단백재암조직대다표체위~삽수평,암방정상조직칙대다표체재-~+수평,NGAL단백재암조직표체현저고우암방정상조직(P<0.01).NGAL mRNA화단백표체여림파결전이、병리류형、ER、HER-2상태명현상관(P<0.05),이여환자년령、종류대소、조직학분급、TNM분기、PR상태무명현상관(P>0.05).결론 NGAL적표체여유선암림파결전이화내분비치료내약등상관,NGAL가능시유선암조기진단、감측복발화판단예후적잠재생물학삼고지표.
Objective To explore the neutrophil gelatinase-associated lipocalin (NGAL) expression in breast cancer and its clinical significance. Methods Forty-seven cases of human breast cancer specimens were collected from the patients who received operation for breast cancer. Semi-quantitative RTPCR and immunohistochemistry were used to detect the expression of NGAL mRNA and protein expression respectively. Results The results of semi-quantitative RT-PCR analysis showed that the level of NGAL mRNA expression in primary breast carcinoma specimens was 0.42 ± 0.19, significantly higher than in the paracancerous tissues (P <0.01 ) ;Immunohistochemistry staining revealed that the degree of NGAL protein expression in primary breast cancer tissues ( average ) was significantly higher than in the paracancerous tissues (average -- + ,P <0.01 ). The high-expression of NGAL mRNA and protein was related to lymph node metastases, pathological type, ER and HER-2 status ( P < 0.05 ). However, there was no significant association with age, tumor size, histologic grade, TNM stage and PR status (P > 0.05 ). Conclusion The expression of NGAL was closely correlate with lymph node metastases and resistance of endocrine therapy, suggesting that NGAL might be a potential biomarker for the early diagnosis, monitoring recurrence and predication of prognosis of human primary breast cancer.
