目的 观察外源性细胞因子白细胞介素( IL)-2、干扰素(IFN)-γ和IL-4单克隆抗体(McAb)、IL-10 McAb在体外能否促使Th2型细胞因子分泌优势向Th1型逆转,抑制胶质瘤细胞生长。方法 体外培养大鼠C6胶质瘤细胞,加入外源性细胞因子共同孵育,光学显微镜观察细胞形态学变化,噻唑蓝(MTT)比色法检测细胞增殖抑制率,逆转录-聚合酶链反应(RT-PCR)检测Th1/Th2类细胞因子表达变化。结果 实验组较对照组细胞体积变小,生长密度低,细胞数目少;实验组与对照组比较细胞增殖抑制率分别为(26.41 ±3.60)%,其A值与对照组比较差异有统计学意义(P<0.05)。对照组细胞只表达Th2类细胞因子,Th1类细胞因子表达缺如,实验组IFN-γ表达增加,IL-4和IL-10表达减弱,但是未见IL-2表达。结论 外源性细胞因子能够抑制脑胶质瘤细胞的生长,促进Th2型细胞因子分泌优势向Th1型逆转。

목적 관찰외원성세포인자백세포개소( IL)-2、간우소(IFN)-γ화IL-4단극륭항체(McAb)、IL-10 McAb재체외능부촉사Th2형세포인자분비우세향Th1형역전,억제효질류세포생장。방법 체외배양대서C6효질류세포,가입외원성세포인자공동부육,광학현미경관찰세포형태학변화,새서람(MTT)비색법검측세포증식억제솔,역전록-취합매련반응(RT-PCR)검측Th1/Th2류세포인자표체변화。결과 실험조교대조조세포체적변소,생장밀도저,세포수목소;실험조여대조조비교세포증식억제솔분별위(26.41 ±3.60)%,기A치여대조조비교차이유통계학의의(P<0.05)。대조조세포지표체Th2류세포인자,Th1류세포인자표체결여,실험조IFN-γ표체증가,IL-4화IL-10표체감약,단시미견IL-2표체。결론 외원성세포인자능구억제뇌효질류세포적생장,촉진Th2형세포인자분비우세향Th1형역전。
Objective To study whether exgenous interleukin ( IL)-2, interferon ( IFN )-γ and IL-4 monoclonal antibody (McAb) and IL-10 McAb can cause the superiority of Th2-type cytokines secretion to the Thl reversion and inhibit the growth. Methods In vitro C6 cells were cuhured in the presence of exogenous IL-2, IFN-γ, IL-4 McAb and IL-10 McAb. The cell morphology changes were observed under the microscopy. The gene expression of Th1/Th2 cytokines was detected by using reverse transcription-polymerase chain reaction ( RT-PCR), and inhibition rate of cell proliferation was measured by using methyl thiazol tetrazolium (MTT) assay. Results In control group, the cells had irregular morphology, the cells were shrinkaged, the volume, number of cells and the cells growth density were reduced obviously as compared with experimental group. The inhibition rate of cells proliferation in control group was (26. 41 ±3.60) % (P < 0. 05 ). RT-PCR result showed the exprimental group had IFN-γ expression, but had no expression of IL-2 and IL-4, and the expression of IL-10 was relatively weakened. Conclusion The experimental results indicated that exogenous Th1-type cytokines and Th2-type cytokines antibody may inhibit glioma cells growth, and may promote the superiority of Th2-type cytokines secretion to the Th1 reversion.
