目的对rhBMP-2在诱导成骨中Collagen I,II及碱性磷酸酶(ALP)的表达进行研究. 方法将含rhBMP-2 0.5 mg的松质骨载体植入55只BALB/c小鼠的右侧股部肌袋内,分别于术后3~21 d共8个时间点取材,采用免疫组织化学方法对新生骨组织中的I,II型胶原进行检测;对ALP活性进行定量分析,观察rhBMP-2在诱导成骨中Collagen I,II及ALP的表达情况. 结果①II型胶原的出现是和成软骨、软骨细胞的出现相伴随的,但是,肥大、变性的软骨细胞并不表达II型胶原; ②作为成骨细胞成熟标志物的I型胶原、ALP在软骨形成期即有表达,但是随着成骨细胞的出现,骨组织的形成I型胶原仍表现为高表达,而ALP的表达则呈下降趋势. 结论在诱导成骨中rhBMP-2促进了Collagen I,II及ALP的表达.
目的對rhBMP-2在誘導成骨中Collagen I,II及堿性燐痠酶(ALP)的錶達進行研究. 方法將含rhBMP-2 0.5 mg的鬆質骨載體植入55隻BALB/c小鼠的右側股部肌袋內,分彆于術後3~21 d共8箇時間點取材,採用免疫組織化學方法對新生骨組織中的I,II型膠原進行檢測;對ALP活性進行定量分析,觀察rhBMP-2在誘導成骨中Collagen I,II及ALP的錶達情況. 結果①II型膠原的齣現是和成軟骨、軟骨細胞的齣現相伴隨的,但是,肥大、變性的軟骨細胞併不錶達II型膠原; ②作為成骨細胞成熟標誌物的I型膠原、ALP在軟骨形成期即有錶達,但是隨著成骨細胞的齣現,骨組織的形成I型膠原仍錶現為高錶達,而ALP的錶達則呈下降趨勢. 結論在誘導成骨中rhBMP-2促進瞭Collagen I,II及ALP的錶達.
목적대rhBMP-2재유도성골중Collagen I,II급감성린산매(ALP)적표체진행연구. 방법장함rhBMP-2 0.5 mg적송질골재체식입55지BALB/c소서적우측고부기대내,분별우술후3~21 d공8개시간점취재,채용면역조직화학방법대신생골조직중적I,II형효원진행검측;대ALP활성진행정량분석,관찰rhBMP-2재유도성골중Collagen I,II급ALP적표체정황. 결과①II형효원적출현시화성연골、연골세포적출현상반수적,단시,비대、변성적연골세포병불표체II형효원; ②작위성골세포성숙표지물적I형효원、ALP재연골형성기즉유표체,단시수착성골세포적출현,골조직적형성I형효원잉표현위고표체,이ALP적표체칙정하강추세. 결론재유도성골중rhBMP-2촉진료Collagen I,II급ALP적표체.
AIM To study expression of collagen I, II and alkaline phosphatase in osteogenesis by rhBMP-2. METHODS Compositions of rhBMP-2 (0.5 mg) and cancellous bone carrier were implanted in the right thigh pouch of 55 BALB/c mice. In order to investigate expression of collagen I, II and alkaline phosphatase in osteogenesis by rhBMP-2, immunohistochemistry analysis was performed 3, 5, 7, 9, 11, 14, 17 and 21 days after the implantation. Meanwhile, alkaline phosphatase activity was measured 7, 14, 21 days after the implantation. RESULTS 1. The appearance of type II collagen was accompanied by that of chondroblasts and chondrocytes, but hypertrophic and degenerative chondrocytes did not express type II collagen. 2. Type I collagen and ALP, osteoblast differentiation markers, were expressed at the early stage of chondrogenesis, but type I collagen was still expressed at higher level, ALP decreased gradually with the appearance of osteoblasts and osteogenesis. CONCLUSION rhBMP-2 might stimulate expression of collagen I, II and alkaline phosphatase in osteogenesis.
