目的 了解坐骨神经切断后远端神经段内转化生长因子β1(TGF-β1)和Ⅲ型胶原蛋白(COL-Ⅲ)的变化规律及相关性.方法 将大鼠右侧坐骨神经切断,分别于术后不同时间段取远端神经,用HE染色、免疫组织化学染色、RT-PCR、Western Blot技术检测远端神经的组织形态学变化、TGF-β1和COL-Ⅲ的表达及含量变化.结果 坐骨神经切断后远端神经呈现纤维化的改变,TGF-β1迅速增加,7d及28 d时呈现双高峰.COL-Ⅲ逐渐降低,至7d时达最低点后逐渐增加,在28d时达到高峰后处于一个高于正常水平的平台期,且相关性分析(R=0.546,P<0.01)提示两者具有相关性.结论 坐骨神经切断后远端神经段内TGF-β1和COL-Ⅲ的生成具有相关性,说明TGF-β1与损伤神经纤维化密切相关.

Objective To investigate the changes and correlation of transforming growth factor β1(TGF-β1)and collagen Ⅲ(COL-Ⅲ)in the distal nerve segment after sciatic nerve transection in rats.Methods The right sciatic nerve of rats was cut,and the distal nerve segments were harvested after different intervals.H&E stain,immunohistochemistry,RT-PCR,Westem-blot techniques were used to detect the morphologic changes of the distal nerve and the expression and content of TGF-β1 and COL-Ⅲ.Results Sciatic nerve transection resulted in fibrosis,rapid increase of TGF-β1 with double peak at 7 and 28 days in the distal nerve segment.COL-Ⅲ gradually reduced to the lowest point at 7 days and then increased gradually and peaked at 28 days at a higher than normal level after the plateau.Correlation analysis(R =0.546,P < 0.01)suggested the two changes were closely related.Condusion TGF-β1 and COL-Ⅲ are correlated in the distal nerve segment after sciatic nerve transection.TGF-β1 is closely related to fibrosis after the nerve damage.
