利用柱前衍生化反相高效液相色谱法检测不同前处理全蚕粉中1-脱氧野尻霉素(1-deoxynojirimycin, DNJ)的提取分离效果,并研究其分离产物对人宫颈癌HeLa细胞的抗肿瘤作用.结果表明,超低温冷冻干燥、热风烘干和真空浓缩煎煮冷冻干燥所得全蚕粉中DNJ的质量分数分别为 0.494 9%、0.383 9%和0.607 6%;冷冻干燥全蚕粉粉碎粒度为0.15、0.25和0.425 mm时,DNJ质量分数分别为0.494 9%、0.321 5%和0.089 5%;用稀酸浸渍及阳离子交换树脂提取与分离全蚕粉中的DNJ效果最佳,分离物中DNJ的质量分数达到1.08%.浓度为 40 mg/mL的全蚕粉分离物对肿瘤细胞的生长抑制率达92.27%,可使肿瘤细胞增殖能力完全丧失.结果提示:采用真空浓缩煎煮结合超低温冷冻干燥、超细粉碎和稀酸浸渍及阳离子交换树脂分离技术,可有效提高全蚕粉中DNJ的提取、分离效率,含DNJ的全蚕粉分离物对HeLa肿瘤细胞的生长具有显著的抑制作用.

By using the pre-column derivatization reverse phase high performance liquid chromatography, the efficiency of 1-deoxynojirimycin (DNJ) isolation from silkworm powder prepared with different pre-treatment methods was investigated. The obtained isolate was examined for their inhibitory effect to HeLa tumor cells. The results showed that, in the ultra-low temperature freeze-dried, heat-dried and vacuum condensation and decoction combining with ultra-low temperature freeze-dried, silkworm powder samples, DNJ content was 0.494 9%, 0.383 9% and 0.607 6% respectively. DNJ content in the freeze-dried samples with particle sizes of 0.15, 0.25 and 0.425 mm was 0.494 9%, 0.321 5% and 0.089 5%, respectively. In the isolate obtained from dilute acid immersion and cation exchange resin, DNJ content was as high as 1.08%. The silkworm powder isolate with concentration of 40 mg/mL had inhibition efficiency of 92.27% to HeLa tumor cells, being able to eliminate tumor cell proliferation completely. The results implied that, vacuum condensation and decoction combining with ultra-low temperature freeze-drying, ultra-fine grinding technology and dilute acid immersion and cation exchange resin could improve the isolation efficiency of DNJ from silkworm powder remarkably, and the silkworm powder isolate containing DNJ had significant inhibitory effect to the growth of HeLa tumor cells.
