目的 探讨金匮肾气丸治疗肾阳虚型中老年男性雄激素部分缺乏的作用机制.方法 取15月龄SD大鼠40只,随机分为模型组和金匮肾气丸低、中、高剂量治疗组,共4组,每组均10只,各组大鼠均给予腹腔注射环磷酰胺混悬液(1ml/kg·d~(-1)),qd,连续5d,造成雄激素部分缺乏模型后,低、中、高剂量组分别给与金匮肾气丸混悬液灌胃,模型组以蒸馏水灌胃,灌胃28d.末次给药24h后处死大鼠,放射免疫法检测血清T水平,免疫组化法测定3β-HSD活性表达.结果 与模型组比较,金匮肾气丸中、高剂量组血清T水平明显升高(P<0.05);金匮肾气丸各组3β-HSD阳性表达均显著增强(P<0.01),以中、高剂量组作用最为显著.结论 金匮肾气丸可通过增强睾酮合成通路中关键酶3β-HSD的活性表达来提高睾酮的合成,从而达到治疗雄激素部分缺乏的目的.
目的 探討金匱腎氣汍治療腎暘虛型中老年男性雄激素部分缺乏的作用機製.方法 取15月齡SD大鼠40隻,隨機分為模型組和金匱腎氣汍低、中、高劑量治療組,共4組,每組均10隻,各組大鼠均給予腹腔註射環燐酰胺混懸液(1ml/kg·d~(-1)),qd,連續5d,造成雄激素部分缺乏模型後,低、中、高劑量組分彆給與金匱腎氣汍混懸液灌胃,模型組以蒸餾水灌胃,灌胃28d.末次給藥24h後處死大鼠,放射免疫法檢測血清T水平,免疫組化法測定3β-HSD活性錶達.結果 與模型組比較,金匱腎氣汍中、高劑量組血清T水平明顯升高(P<0.05);金匱腎氣汍各組3β-HSD暘性錶達均顯著增彊(P<0.01),以中、高劑量組作用最為顯著.結論 金匱腎氣汍可通過增彊睪酮閤成通路中關鍵酶3β-HSD的活性錶達來提高睪酮的閤成,從而達到治療雄激素部分缺乏的目的.
목적 탐토금궤신기환치료신양허형중노년남성웅격소부분결핍적작용궤제.방법 취15월령SD대서40지,수궤분위모형조화금궤신기환저、중、고제량치료조,공4조,매조균10지,각조대서균급여복강주사배린선알혼현액(1ml/kg·d~(-1)),qd,련속5d,조성웅격소부분결핍모형후,저、중、고제량조분별급여금궤신기환혼현액관위,모형조이증류수관위,관위28d.말차급약24h후처사대서,방사면역법검측혈청T수평,면역조화법측정3β-HSD활성표체.결과 여모형조비교,금궤신기환중、고제량조혈청T수평명현승고(P<0.05);금궤신기환각조3β-HSD양성표체균현저증강(P<0.01),이중、고제량조작용최위현저.결론 금궤신기환가통과증강고동합성통로중관건매3β-HSD적활성표체래제고고동적합성,종이체도치료웅격소부분결핍적목적.
Objectives To study the effect of Jingkui Shenqi Pill on serum testosterone and the activity expression of 3β-HSD of rats with partial androgen deficiency and explore its phamacological mechanism.Methods Total of 40 15-month-old SD rats were randomly divided into four groups such as model group, low dosage Jinkui Shenqi Pill group, middle dosage Jinkui Shenqi Pill group and high dosage Jinkui Shenqi Pill group.There were 10 rats in each group.Cyclophosphamide was injected into the abdominal cavity of all rats (20mg/kg·d~(-1), 5 continuous days) for establishment of rat model with partial androgen deficiency.Then, they were administrated different dose Jinkui Shenqi Pill by gastrogavage for 28 days.The level of T was measured by radioimmunoassay, and the expression of 3β-HSD was detected with immunohistochemistry.Results Compared with that of rats in the model group, the level of serum T of rats in the middle and high dosage groups was increased significantly(P<0.05); The higher expression of 3β-HSD was found in all the treated groups (P<0.01), especially in the middle and high treated groups.Conclusion Jinkui Shenqi Pill might upregulate the activity expression of 3β-HSD, and further enhance the synthesis of testosterone for treatment of androgen deficiency.
