[目的] 寻求甜叶悬钩子组织培养中的最佳抗褐化剂.[方法] 以甜叶悬钩子的茎和叶为外植体,以MS为基本培养基,加入不同浓度的抗褐化剂(20% Na2S2O3、Vc和AC),研究不同抗褐化剂对甜叶悬钩子组织培养的影响.[结果] AC和Vc都能对不同外植体起到抗褐化作用,在加入20% Na2S2O3的培养基上,外植体的褐化程度与Na2S2O3的浓度成正比,培养基中20% Na2S2O3的添加量达20 ml时,外植体褐化最为迅速;在加入Vc的培养基上,随着Vc添加量的增加,培养基变成液体或半固体培养基,接入的外植体容易死亡;在加入AC的培养基上,AC的添加量为5 g/L时,外植体的褐化率可控制在5%以下.抗褐化控制处理后,可以在诱导愈伤组织的培养基上培养出大量的愈伤组织.[结论] AC是较为理想的抗褐化剂,用量为5 g/L.

[목적] 심구첨협현구자조직배양중적최가항갈화제.[방법] 이첨협현구자적경화협위외식체,이MS위기본배양기,가입불동농도적항갈화제(20% Na2S2O3、Vc화AC),연구불동항갈화제대첨협현구자조직배양적영향.[결과] AC화Vc도능대불동외식체기도항갈화작용,재가입20% Na2S2O3적배양기상,외식체적갈화정도여Na2S2O3적농도성정비,배양기중20% Na2S2O3적첨가량체20 ml시,외식체갈화최위신속;재가입Vc적배양기상,수착Vc첨가량적증가,배양기변성액체혹반고체배양기,접입적외식체용역사망;재가입AC적배양기상,AC적첨가량위5 g/L시,외식체적갈화솔가공제재5%이하.항갈화공제처리후,가이재유도유상조직적배양기상배양출대량적유상조직.[결론] AC시교위이상적항갈화제,용량위5 g/L.
[Objective] The purpose was to seek for optimum anti-browning agent in the tissue culture of Rubus suavissimus S.Lee.[Method] With stems and leaves of R.suavissimus as explants, MS basic medium was used as the media by adding different concn.of anti-browning agent (20% Na2S2O3, Vc and AC) so as to research the effect of different anti-browning agent on tissue culture of R.suavissimus.[Result] AC and Vc had anti-browning effect on different explants.On the medium with 20% Na2S2O3, the browning extent of explants was proportional to the concn.of Na2S2O3.When the addition of 20% Na2S2O3 amounted to 20 ml, the browning of the explant was the fastest.On the medium with Vc, with the increasing addition of Vc, the medium got into a liquid or semi-solid medium and the explants died easily.On the medium with AC, when the addition of AC was 5 g/L, the browning rate of the explants could be controlled below 5%.A large number of callus could be induced on the culture medium after the anti-browning control treatment.[Conclusion]AC was the best anti-browning agent and dosage was 5 g/L.
