采用CuCl_2处理小麦幼苗的方法,研究重金属元素铜对小麦幼苗生理生化及蛋白组分影响.结果表明:随着环境中Cu~(2+)浓度的增加小麦苗株高呈显著降低趋势,各浓度处理的苗高均极显著低于对照,5 mmol/L处理较对照降低了54.47%;叶绿素含量呈现降-升-降的趋势,但均小于对照;可溶性蛋白含量分别比对照降低了0.68~1.23 mg/g,但差异不显著.利用双向电泳技术分析了1 mmol/L Cu~(2+)处理后叶绿体蛋白质组分变化,结果表明,有17个蛋白质斑点发生较明显变化,其中7个斑点缺失,1个斑点增加,另有9个蛋白质斑点表达量降低.

The aim of studying the effects of heavy metal copper which is one of the heavy metal pollutants on the physiological, biochemical characteristics and chloroplast proteome of the wheat seedlings is to explore its influence on the growth of wheat seedlings and chloroplast proteome. Treat the wheat seedlings with CuCl_2 for 5 days, measure the length of seedlings and determine the content of chlorophyll and soluble protein, the results indicated that with the concentration of Cu~(2+) increased, the height of wheat seedlings significantly decreased, the height of seedlings under each concentration was significantly lower than the control,54.47% decreased with the concentration of 5 mmol/L in contrast to the control;the chlorophyll content showed the trend of decrease-increase-decrease,but all less than the control;the soluble protein con-tent decreased 0.68 - 1.23 mg/g in contrast to the control with no significant distinction. Analyze the change of chloro-plast proteome by two-dimensional electrophoresis (2-DE) with the Cu~(2+) concentration of 1 mmol/L, the result showed that 17 protein spots significantly changed,seven spots disappeared,one spot appeared,and nine spots down-expressed.
