利用6-BA、IBA、pH和培养基种类、琼脂浓度、KCl浓度各三个因素三水平进行L_9(3~4)正交实验,研究各因素对铁皮石斛原球茎分化和生根的影响.对实验统计通过极差分析及方差分析,结果表明:在基本培养基N_6中,当6-BA的浓度为2 mg/L、IBA浓度为0.2 mg/L、pH值为5.8时,原球茎最易于分化成丛生小苗;影响原球茎分化程度的因素依次为IBA浓度>6-BA浓度>pH值.铁皮石斛组培丛生苗生根的最优配方是:N_6+2.5 mol/L KCl+4 g/L 琼脂;影响诱导生根的主要因素是KCl浓度,其次是琼脂浓度,最后是基本培养基种类.

To study the effects of different factors on protocorm differentiation and root induction of Dendrobium officinale, two groups of three kinds of factors on three levels were used by orthogonal design. The data were analyzed with range analysis and variance analysis. Results showed that the optimal media to induce cluster sheet from protocorm was N_6 medium with 6-BA 2 mg/L and IBA 0.2 mg/L and the pH 5.8. The efficiency of the three factors was in sequence of IBA> 6-BA> pH. On the other hand, N_6 medium with KCl 2.5 mol/L and agar 4 g/L was suitable for root induction of Dendrobium officinale. The main factor which influenced root induction was KCl concentration, and the others minor factors were agar concentration and type of basic medium.
