目的 筛选直肠癌放疗前后差异表达蛋白,发现辐射敏感相关蛋白.方法 采用荧光差异显微凝胶电泳(DIGE)技术分离并筛选的Cy3、Cy5及Cy2荧光素标记的放疗前后的直肠癌组织的差异表达的蛋白质,用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术进行鉴定并分析.结果 共筛选出20个蛋白点,通过MALDI-MS进行鉴定后,有17个蛋白质点成功,在蛋白质库中找到匹配蛋白18个,发现上调蛋白12个,分别为DHE3、DHE4、VIME、CYHR1、CH60、ACTG、MBE2N、ACTS、G3P、ANXA2、MYI6、RLA2;下调蛋白6个,分别为PMR9、PDIA3、FIBB、CATD、LEG3、TXND.结论 蛋白质组学在直肠癌的辐射相关蛋白研究中,能发现更多与辐射敏感相关的蛋白.

Objective To screen the differential expression proteins of rectal cancer after radiotherapy and find out radiosensitivity-related proteins.Methods The cancer tissues were collected before and after radiotherapy,and examined by the fluorescence differential in-gel electrophoresis (DIGE) technique after labeled with CyDye DIGE fluors Cy3,Cy5 and Cy2.The protein spots detected by DIGE were identified by MALDI-TOF-MS.Results Intensity changes of 20 spots were detected with statistically significant difference.There were 17 protein spots identified by MALDI-MS successfully,and 18 matching proteins were found in protein bank,including 12 proteins which were up-regulated ( DHE3,DHE4,VIME,CYHR1,CH60,ACTG,MBE2N,ACTS,G3P,ANXA2,MYI6,RLA2) and 6 proteins which were down-regulated (PMR9,PDIA3,FIBB,CATD,LEG3,TXND).Conclusion Protemics can be applied widely in the study about radio-related proteins of rectal cancer.
