目的研究载有外源性TGF-β1复合材料,在体外对小鼠成骨样细胞系(MC3T3-E1)生物活性的影响。方法将MC3T3-E1接种于96孔板中,分别加入不同浓度TGF-β1溶液(0.2ng/ml,1ng/ml,5ng/ml)和FHA、FA悬浮液的混合溶液,并设置对照组,分别培养1d、3d、5d,用酶联免疫检测仪测定OD值以检测细胞活力。结果各组细胞在3d时均出现成倍增殖,除空白组外5d时均增殖减弱。浓度为1ng/ml TGF-β1比0.2ng/ml和5ng/ml具有更强的促成骨细胞增殖作用。结论 TGF-β1与FHA、FA的混合溶液可改变成骨细胞增殖周期,使之提前出现增殖高峰,且1ng/ml TGF-β1溶液对成骨细胞具有最强的促增殖作用。

목적연구재유외원성TGF-β1복합재료,재체외대소서성골양세포계(MC3T3-E1)생물활성적영향。방법장MC3T3-E1접충우96공판중,분별가입불동농도TGF-β1용액(0.2ng/ml,1ng/ml,5ng/ml)화FHA、FA현부액적혼합용액,병설치대조조,분별배양1d、3d、5d,용매련면역검측의측정OD치이검측세포활력。결과각조세포재3d시균출현성배증식,제공백조외5d시균증식감약。농도위1ng/ml TGF-β1비0.2ng/ml화5ng/ml구유경강적촉성골세포증식작용。결론 TGF-β1여FHA、FA적혼합용액가개변성골세포증식주기,사지제전출현증식고봉,차1ng/ml TGF-β1용액대성골세포구유최강적촉증식작용。
Objective Research the biological activity of exogenous TGF-β1 composite materials in vitro and its effect on signal expression of osteoblastic cell(MC3T3-E1). Methods MC3T3-E1 was seeded in 96-well plates,and added to mixed solution with different concentrations of TGF-β1 solution(0.2,1,5 ng / ml)and FHA,FA suspension, and control groups were set up. The groups were cultured 1d,3d,5d,and then OD values measured by enzyme-linked immunosorbent assay to detect cell viability. Results Cells of each group proliferated visibly in 3d,and decreased slightly in 5d except the blank,the concentration of 1ng/ml TGF-β1 had stronger promotion than 0.2ng/ml and 5ng/ml on cell proliferation. Conclusion Mixed solution with TGF-β1 and FHA、FA suspension can alter the proliferation cycle of osteoblasts,and advance the peak of proliferation. 1ng/ml TGF-β1 solution has the strongest effect on the proliferation of osteoblasts.
