目的:检测碘131(131I)对转染了人钠/碘转运体(hNIS)基因的人结肠癌细胞(SW480细胞)的杀伤作用。方法:将SW480细胞分为转染组、空白质粒转染组(空白质粒组)和空白对照组,行体外摄131I试验、高氯酸盐抑制实验,及AnnexinV-FITC/PI双染行流式细胞仪检测细胞凋亡情况。结果:转染组与空白对照组及空白质粒组比较,摄131I能力均增高8倍;高氯酸盐可以有效地抑制转染组的摄131I能力;AnnexinV-FITC/PI双染后流式细胞仪检测结果显示,转染组凋亡指数高于空白对照组和空白质粒组。结论:131I对转染hNIS-SW480细胞具有显著杀伤效果。
目的:檢測碘131(131I)對轉染瞭人鈉/碘轉運體(hNIS)基因的人結腸癌細胞(SW480細胞)的殺傷作用。方法:將SW480細胞分為轉染組、空白質粒轉染組(空白質粒組)和空白對照組,行體外攝131I試驗、高氯痠鹽抑製實驗,及AnnexinV-FITC/PI雙染行流式細胞儀檢測細胞凋亡情況。結果:轉染組與空白對照組及空白質粒組比較,攝131I能力均增高8倍;高氯痠鹽可以有效地抑製轉染組的攝131I能力;AnnexinV-FITC/PI雙染後流式細胞儀檢測結果顯示,轉染組凋亡指數高于空白對照組和空白質粒組。結論:131I對轉染hNIS-SW480細胞具有顯著殺傷效果。
목적:검측전131(131I)대전염료인납/전전운체(hNIS)기인적인결장암세포(SW480세포)적살상작용。방법:장SW480세포분위전염조、공백질립전염조(공백질립조)화공백대조조,행체외섭131I시험、고록산염억제실험,급AnnexinV-FITC/PI쌍염행류식세포의검측세포조망정황。결과:전염조여공백대조조급공백질립조비교,섭131I능력균증고8배;고록산염가이유효지억제전염조적섭131I능력;AnnexinV-FITC/PI쌍염후류식세포의검측결과현시,전염조조망지수고우공백대조조화공백질립조。결론:131I대전염hNIS-SW480세포구유현저살상효과。
Objective: To detect the killing effect of colon cancer cells (SW480) by iodine 131 (131I). Methods: Cell apoptosis was detected when taken in131I in vitro test and perchlorate suppression experiments and AnnexinV-FITC/PI double dye lfow cytometry instrument.Results: Stably-hNIS-gene-expressing cell line was about 8 times higher than the control group and blank plasmid group in 24 h iodine uptake capacity. Per-chlorate inhibition tests showed that the iodine uptake of hNIS-SW480 was the hNIS gene’s function. AnnexinV-FITC/PI double dye lfow cytometry showed apoptosis index in the transfection group was higher than that of the control group and blank plasmid group. There were signiifcant differences that radioactive iodine SW480 cells transfected with better killing ability under 100 μCi131I in 24 h.Conclusion: 131I has signiifcant cell killing effect on SW480 cells transfected hNIS.
