为了从大叶南五味子多糖(KPS)中获得单体,将其进行分级与纯化。用乙醇为沉淀剂对KPS进行分级,Sephadex柱色谱梯度洗脱分离各级多糖,蒽酮-硫酸比色法测定糖含量及洗脱液吸光度值。乙醇分级沉淀结果显示KPS分为Ⅰ~Ⅴ级分,糖含量分别为76.51%、78.36%、95.13%、92.41%、88.35%;Sephadex柱色谱分离结果显示,KPSⅡ、Ⅲ、Ⅴ中所含多糖分子量基本一致,为纯一多糖;KPSⅠ、Ⅳ中含有不同分子量多糖,其中KPSⅠ-1、KPSⅡ-1、KPSⅢ-1、KPSⅣ-6、KPSⅤ-1均为纯多糖。分级纯化的大叶南五味子多糖单体为其化学结构的分析奠定基础。

In order to obtain the monomer of polysaccharide, Kadsura marmorata polysaccharides (KPS) was separated and purified. KPS was fractionally precipitated by the different concentration ethanol. The polysaccharide monomers were separated through Sephadex column chromatography and NaHCO3 solution gradient elution. The content of sugar and absorbance value of eluant component was detected by anthrone-vitriol colorimetric method. The results of ethanol fractionally precipitation showed that KPS was separated five fractional polysaccharides (KPSⅠ-Ⅴ), and the content of five sugars was 76.51%, 78.36%, 95.13%, 92.41%, 88.35%, respectively. The results of separation by Sephadex column chromatography showed that KPSⅡ, KPSⅢand KPSⅤcontained the single molecular weight polysaccharide, respectively. However, KPS Ⅰ and KPS Ⅳ contained the different molecular weight polysaccharides, respectively. Those polysaccharide monomers, which included KPSⅠ-1, KPSⅡ-1, KPSⅢ-1, KPSⅣ-6 and KPSⅤ-1, were separated by Sephadex colum. This work will lay a foundation for the further research chemical structure analysis of KPS.
