Tympanosclerosis%OPG%RANKL
目的检测OPG和RANKL在大鼠鼓室硬化模型中的表达水平,探讨OPG/RANKL系统在大鼠鼓室硬化发病机制中的作用。方法30只健康的成年雄性SD大鼠随机分为两组,造模组20只大鼠,其右耳鼓室内注射浓度为1×108 CFU/ml的肺炎链球菌液进行造模;对照组10只大鼠,其右耳不干预作为空白对照。在造模后第8周处死大鼠,收集造模组大鼠右耳中有鼓室硬化形成的听泡标本作为鼓室硬化组和对照组大鼠右耳的听泡标本作为对照组,进行免疫组化染色。结果对照组的10只大鼠全部存活到造模后第8周,右侧10耳均未发生鼓室硬化;造模组共有17只大鼠存活至造模后第8周,其右侧17耳中共有13耳发生鼓室硬化。鼓室硬化组鼓室黏膜中的OPG表达水平明显高于对照组(P<0.05),鼓室硬化组鼓室黏膜中的OPG/RANKL比值明显高于对照组(P<0.05)。结论OPG/RANKL系统可能在大鼠鼓室硬化的发病过程中发挥调节作用,OPG/RANKL比值增加可能与大鼠鼓室硬化的形成有关。

목적검측OPG화RANKL재대서고실경화모형중적표체수평,탐토OPG/RANKL계통재대서고실경화발병궤제중적작용。방법30지건강적성년웅성SD대서수궤분위량조,조모조20지대서,기우이고실내주사농도위1×108 CFU/ml적폐염련구균액진행조모;대조조10지대서,기우이불간예작위공백대조。재조모후제8주처사대서,수집조모조대서우이중유고실경화형성적은포표본작위고실경화조화대조조대서우이적은포표본작위대조조,진행면역조화염색。결과대조조적10지대서전부존활도조모후제8주,우측10이균미발생고실경화;조모조공유17지대서존활지조모후제8주,기우측17이중공유13이발생고실경화。고실경화조고실점막중적OPG표체수평명현고우대조조(P<0.05),고실경화조고실점막중적OPG/RANKL비치명현고우대조조(P<0.05)。결론OPG/RANKL계통가능재대서고실경화적발병과정중발휘조절작용,OPG/RANKL비치증가가능여대서고실경화적형성유관。
Objective The aim of this study was to investigate expression of OPG and RANKL in the tympanic mucosa of experimental tympanosclerosis and the regulative effects of the OPG/RANKL system on the pathogenesis of tympanosclero-sis in rats. Methods A total of 30 healthy adult male Sprague-Dawley rats were randomly divided into a modeling (n=20) and a control group (n=10). Tympanosclerosis was induced by inoculating 0.05 mL of solution containing 1×108 CFU/ml strepto-coccus pneumonia into the right middle ear in the 20 rats in the modeling group. Rats in the control group received no inocula-tion. All animals were sacrificed at the end of 8 weeks. The right tympanic bulla was harvested and stained immunohistochem-ically to detect OPG and RANKL expression. Result None of the 10 rats in the control group showed tympanosclerosis. A to-tal of 17 rats in the modeling group survived after eight weeks and 13 showed tympanosclerosis. The level of OPG expression in tympanic mucosa was significantly higher in the modeling group than in the control group (P<0.05).The OPG and RANKL expression ratio in tympanic mucosa was also significantly higher in the modeling group than in the control group (P<0.05). Conclusion OPG/RANKL system may play a regulative role in the development of tympanosclerosis in rats. The increased OPG to RANKL ratio may be related to the development of tympanosclerosis in rats.
