目的：探讨 NVP-BEZ235对卵巢癌细胞 SKOV3的增殖抑制及体内、体外转移的抑制作用。方法对贴壁生长的卵巢癌细胞SKOV3进行体外培养及传代,采用Cell Count Kit-8检测不同浓度NVP-BEZ235(125、250、500、1000 nmol/mL)处理SKOV3细胞48 h后的增殖抑制效应。选取无增殖抑制的浓度125 nmol/mL处理细胞,Transwell法检测体外侵袭变化,斑马鱼移植癌细胞模型检测体内侵袭变化。结果 NVP-BEZ235能显著抑制卵巢癌细胞SKOV3体外增殖, Transwell法检测体外侵袭能力明显减弱, NVP-BEZ235显著抑制 SKOV3细胞在斑马鱼体内转移。结论 NVP-BEZ235对于卵巢癌细胞SKOV3具有抑制增殖,减少体外体内侵袭,具有成为卵巢癌治疗药物的可能。
목적：탐토 NVP-BEZ235대란소암세포 SKOV3적증식억제급체내、체외전이적억제작용。방법대첩벽생장적란소암세포SKOV3진행체외배양급전대,채용Cell Count Kit-8검측불동농도NVP-BEZ235(125、250、500、1000 nmol/mL)처리SKOV3세포48 h후적증식억제효응。선취무증식억제적농도125 nmol/mL처리세포,Transwell법검측체외침습변화,반마어이식암세포모형검측체내침습변화。결과 NVP-BEZ235능현저억제란소암세포SKOV3체외증식, Transwell법검측체외침습능력명현감약, NVP-BEZ235현저억제 SKOV3세포재반마어체내전이。결론 NVP-BEZ235대우란소암세포SKOV3구유억제증식,감소체외체내침습,구유성위란소암치료약물적가능。
Objective To investigate the inhibition effect of NVP-BEZ235 on proliferation in vitro and meta-static capacity of SKOV3 cell in vitro and in vivo. Methods Culture and passage the SKOV3 in vitro,assess the differ-ent doses (125,250,500,1 000 nmol/mL) NVP-BEZ235 inhibition effection on proliferation of SKOV3 by cell count Kit-8 after 48 hours. The metastatic capacity of SKOV3 were evaluated by transwell chamber assay in vitro and a ze-brafish xenograft model in vivo. Results NVP-BEZ235 inhibited the proliferation of SKOV3 cancer cells. NVP-BEZ235 inhibited the metastasis of SKOV3 in transwell chamber assay in vitro and Zebrafish xenograft in vivo. Conclu-sion NVP-BEZ235 can suppress the proliferation and invasiveness of ovarian cancer cells,and it might be a candidate treatment for ovarian cancer.
