从浙江省某疑似鸭病毒性肝炎的发病番鸭群分离到1株鸭肝炎病毒(Duck hepatitis virus,DHV)CH12株,分离毒株经动物回归试验、ELD50测定、中和试验等确定为DHV-1(基因A型DHV)。通过RT-PCR方法对DHV CH12株的全基因组序列进行分段扩增并测序。结果表明,CH12株的全基因组长度为7690 bp(不包括PolyA),包括626 bp的5′UTR,6747 bp的ORF和317 bp的3′UTR。CH12株各基因均与基因A型DHV毒株的序列相似性较高,与基因B型和基因C型DHV毒株的序列相似性较低。全基因组核苷酸、多聚蛋白、VP0、VP3、VP1、2C和3D基因的进化分析均显示,CH12株均与基因A型DHV在同一进化分支上,亲缘关系较近,属于基因A型DHV。

Duck hepatitis virus (DHV) strain CH12 was isolated from a diseased domestic Muscovy duckling in Zhejiang Province and characterized in disease reproduction in host animals, ELD50 determination and duck embryo neutralization. The strain CH12 was identified as DHV-1 (genotype A). The complete genome sequence of the strain CH12 was amplified in RT-PCR. The full length of its genome was 7690 nt, including 626 nt of 5' untranslated region (UTR), an open reading frame of 6747 nt and 317 nt of 3' untranslated region. The pairwise percent identity of the genome sequences between strain CH12 and other genotype A strains were higher than that between strain CH12 and genotype B or genotype C strains. Phylogenetic and evolutionary analysis on complete genome, polyprotein, VP0, VP3, VP1, 2C and 3D indicated that strain CH12 had a very close relationship with DHV genotype A.
