目的:建立大鼠宫腔粘连模型并检测大鼠血液和组织中核转录因子κb( nuclear transcription factor kappa B ,NF-κb)的含量,探讨NF-κb与宫腔粘连的关系。方法用苯酚胶浆法建立宫腔粘连动物模型,HE染色观察病理学变化,免疫组化染色法检测大鼠子宫内膜NF-κb亚基p65蛋白质的表达,Real-time PCR方法检测NF-κb p65 mRNA的表达,ELISA双抗夹心法检测血液中NF-κb p65蛋白质的变化。结果 NF-κb亚基p65蛋白质主要定位于细胞核,在宫腔粘连组的表达量明显高于正常组和假手术组( P<0.05),正常组和假手术组间无差异( P>0.05);NF-κb p65 mRNA在宫腔粘连组的表达量明显高于正常组和假手术组( P<0.05),正常组和假手术组无差异(P>0.05);血液中NF-κb亚基p65蛋白质的表达量在宫腔粘连组高于正常组和假手术组( P<0.05),正常组和假手术组间无差异( P>0.05)。结论宫腔粘连的大鼠,NF-κb亚基p65的表达量明显增高,提示NF-κb介导的信号通路在宫腔粘连的发病机制中起重要作用。

목적:건립대서궁강점련모형병검측대서혈액화조직중핵전록인자κb( nuclear transcription factor kappa B ,NF-κb)적함량,탐토NF-κb여궁강점련적관계。방법용분분효장법건립궁강점련동물모형,HE염색관찰병이학변화,면역조화염색법검측대서자궁내막NF-κb아기p65단백질적표체,Real-time PCR방법검측NF-κb p65 mRNA적표체,ELISA쌍항협심법검측혈액중NF-κb p65단백질적변화。결과 NF-κb아기p65단백질주요정위우세포핵,재궁강점련조적표체량명현고우정상조화가수술조( P<0.05),정상조화가수술조간무차이( P>0.05);NF-κb p65 mRNA재궁강점련조적표체량명현고우정상조화가수술조( P<0.05),정상조화가수술조무차이(P>0.05);혈액중NF-κb아기p65단백질적표체량재궁강점련조고우정상조화가수술조( P<0.05),정상조화가수술조간무차이( P>0.05)。결론궁강점련적대서,NF-κb아기p65적표체량명현증고,제시NF-κb개도적신호통로재궁강점련적발병궤제중기중요작용。
Objective To build rat model of intrauterine adhesions ,to detect the expression of NF -κb p65 in uterine tissue and plasma , and to explore the relationship between NF -κb and intrauterine adhesions .Methods Rat intrauterine adhesions model was set up by phenol mucilage .Histologic changes of uterine were monitored by hematoxylin eosin , NF-κb p65 protein expression of semi -quantitative and location in the rat endometrium were assessed by immunohistochemical staining .The changes of NF -κb p65 mRNA expression were assayed by real -time PCR, NF-κb p65 protein expression in the blood was detected by ELISA .Results The NF-κb p65 protein was located in the nucleus , the expression of NF-κb p65 protein of the intrauterine adhesions group was significantly higher than that of normal group and control group (P<0.05), no significant difference was found between normal group and control group ( P >0.05 );The p65 mRNA of NF -κb of the intrauterine adhesions group expressed significantly higher than that of normal group and control group (P<0.05), and no significant difference was found between normal group and control group ( P>0.05 ); The expression of NF -κb in blood of intrauterine adhesions group were significantly higher than that of normal group and control group (P<0.05),and no significant differences <br> was found between the two groups ( P >0.05 ).Conclusion The expression of NF -κb in the rat intrauterine adhesions model increased obviously which indicated NF -κb plays an important role in intrauterine adhesion as mediated inflammatory factors .
