蛹虫草中含有多种生理活性物质,是中国名贵中药材之一。前期试验发现其发酵液中含有纤溶酶,具有开发成一种新型溶栓药物的价值。以试验室保存的8株蛹虫草为试验菌株,以纤溶酶酶活为指标,进行液体深层培养,确定以C-5作为诱变出发菌株,对其进行原生质体紫外诱变处理,筛选得到31株抗制霉素突变株,抗性菌株以纤溶酶酶活为指标进行液体深层发酵,得到8株纤溶酶产量高于出发菌株15.00%以上的突变株,并从中筛选出3株遗传稳定性良好的正突变株CY-8, CY-18, CY-25,其纤溶酶产量分别比出发菌株提高了28.58%,28.22%,21.78%。

The fungus Cordyceps militaris one of the famous Chinese traditional medicinal mushroom and fibrinolytic enzyme is contained in its fermentation liquid, which is expected to be a new thrombolytic drug. To obtain the highest activity of fibrinolytic enzyme, the submerged liquid fermentation of eight Cordycps militaris strains from our lab are studied. C-1 as original strain, its protoplast is mutated, and 31 nystatin-resistant mutants are obtained. All mutants are second screened through repeated submerged, 8 fibrinolytic enzyme higher 15.00% of mutant and then 3 positive mutants with highly genetic stability, CY-8, CY-18 and CY-25 are obtained. The productivity of the fibrinolytic enzyme of positive mutants increased by 28.58%, 28.22%and 21.78%respectively compared with that of original strain.
