目的:通过观察乌龙丹对胶原诱导性类风湿关节炎模型大鼠的影响,探讨乌龙丹抗类风湿关节炎的作用及其机制。方法:对Ⅱ型胶原诱导性大鼠建立类风湿关节炎模型,通过测定关节炎指数积分、关节肿胀率变化情况,评价乌龙丹的治疗作用,并以Elisa法检测血清及滑膜液中白细胞介素-1β、白细胞介素-6、转化生长因子-β1、白细胞介素-17F、基质金属蛋白酶-3表达情况,分析并探讨其作用机制。结果:乌龙丹各治疗组可明显降低模型大鼠关节炎指数积分,血清及滑液中白细胞介素-1β、白细胞介素-6、转化生长因子-β1、白细胞介素-17F、基质金属蛋白酶-3均有不同程度下降,其中以中剂量组下降最为显著。结论:乌龙丹对胶原诱导性类风湿关节炎模型大鼠关节炎具有良好的治疗作用,其机制可能是通过调节关节滑液及血清中白细胞介素-1β、转化生长因子-β1含量,抑制了CD4+T细胞向Th17细胞分化,从而阻断了白细胞介素-1β、白细胞介素-17F、基质金属蛋白酶-3等因子的生物学效应而达到阻止或延缓类风湿关节炎炎症和关节破坏的作用。

목적:통과관찰오룡단대효원유도성류풍습관절염모형대서적영향,탐토오룡단항류풍습관절염적작용급기궤제。방법:대Ⅱ형효원유도성대서건립류풍습관절염모형,통과측정관절염지수적분、관절종창솔변화정황,평개오룡단적치료작용,병이Elisa법검측혈청급활막액중백세포개소-1β、백세포개소-6、전화생장인자-β1、백세포개소-17F、기질금속단백매-3표체정황,분석병탐토기작용궤제。결과:오룡단각치료조가명현강저모형대서관절염지수적분,혈청급활액중백세포개소-1β、백세포개소-6、전화생장인자-β1、백세포개소-17F、기질금속단백매-3균유불동정도하강,기중이중제량조하강최위현저。결론:오룡단대효원유도성류풍습관절염모형대서관절염구유량호적치료작용,기궤제가능시통과조절관절활액급혈청중백세포개소-1β、전화생장인자-β1함량,억제료CD4+T세포향Th17세포분화,종이조단료백세포개소-1β、백세포개소-17F、기질금속단백매-3등인자적생물학효응이체도조지혹연완류풍습관절염염증화관절파배적작용。
Objective:To explore the effect and mechanism of Wulongdan on rheumatoid arthritis through observing the influence of wulongdan on rat model with collagen-induced arthritis.Methods:The rat model with rheumatoid arthritis was established by collagen type II,evaluating therapeutic effect of wulongdan by measuring arthritis index score and changes of joint swelling rate and analyzing and exploring its mechanism by measuring with elisa method the expression of interleukin-1β in serum and synovial fluid,interleukin-6,transforming growth factor-β1,interleukin-17F and matrix metalloproteinase-3.Results:Wulongdan in the treatment group could obviously reduce the arthritis index score of rat model,and at the same time interleukin-1β in serum and synovial fluid,interleukin-6,transforming growth factor-β1,interleukin-17F and matrix metalloproteinase-3 decreased in various degrees,among which above factors in the middle dose group decreased significantly.Conclusion:Wulongdan had good therapeutic effect on collagen-induced rheumatoid arthritis in rats,whose mechanism may be through the regulation of interleukin-1βin serum and synovial fluid,interleukin-6,transforming growth factor-β1,interleukin-17F and matrix metalloproteinase-3 to prevent or delay the rheumatoid arthritis and joint destruction.
