目的:研究Pax1在乳癌组织中的表达及其对乳癌MDA-MB-231细胞生物学行为的影响。方法:采用Western blot检测200乳腺原发癌和癌旁组织中Pax1蛋白的表达。采用化学合成针对Pax1基因的两个小干扰RNA ( siRNA-Pax1-1和siRNA-Pax1-2)下调该基因的表达,应用MTT和Transwell实验检测其对MDA-MB-231细胞生物学行为的影响。结果:Pax1在乳腺原发癌组织中的表达低于相应的癌旁组织,沉默Pax1表达可促进乳癌细胞增殖、迁移和侵袭( F=52.413和72.741,P<0.001)。结论:Pax1有望成为乳癌早期诊断和预测预后的生物分子标志物。

목적:연구Pax1재유암조직중적표체급기대유암MDA-MB-231세포생물학행위적영향。방법:채용Western blot검측200유선원발암화암방조직중Pax1단백적표체。채용화학합성침대Pax1기인적량개소간우RNA ( siRNA-Pax1-1화siRNA-Pax1-2)하조해기인적표체,응용MTT화Transwell실험검측기대MDA-MB-231세포생물학행위적영향。결과:Pax1재유선원발암조직중적표체저우상응적암방조직,침묵Pax1표체가촉진유암세포증식、천이화침습( F=52.413화72.741,P<0.001)。결론:Pax1유망성위유암조기진단화예측예후적생물분자표지물。
To investigate the expression of paired boxed gene 1 ( Pax1) gene in breast cancer tissue and to ex-plore the influence on proliferation status , migration ability and metastatic ability of breast cancer cells MDA-MB-231 . Methods:Western blot were used to examine the expression of Pax 1 in samps, including 200 normal tissues and 200 pri-mary tumors.MTT and transwell were used to analyze the ability of proliferation , migration, and invasion in Pax1 siRNA-transfected MDA-MB-231 cells.Results: The expression of Pax1 was lower in breast cancer tissues than other tissues . Furthermore, knockdown of Pax1 expression resulted in increased proliferation and invasion in MDA-MB-231 cells in vitro (F=52.413 and 72.741,P<0.001).Conclusion:The expression of Pax1 gene shows and obvious downgraded in breast cancer tissue.Low expression of Pax1 gene can increase apeptosis and proliteration of MDA-MB-231 cells, which may pro-vide a strategy for blocking breast cancer and progression .
