目的:主要探讨氯沙坦对5/6肾切除慢性心衰模型大鼠心血管的影响。方法:雄性SD大鼠30只随机分为假手术组(Sham)、病理组(Nx)和氯沙坦治疗组(Lst)。 Nx组和Lst组行5/6肾切除术,Lst组于术后第7周初于饮水中连续两周给予氯沙坦(10 mg·kg-1·d-1)治疗。8周末处理所有大鼠,测定血流动力学参数、心指数、血管舒张度、血清肌酐、尿素氮以及心肌和血管组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果:与Nx组比较,Lst组大鼠LVEDP降低,LVSP、LVDP绝对值升高,心指数降低,血管舒张度增大,血清中肌酐、尿素氮降低,各组织中MDA含量降低,SOD活性升高,结果有显著性差异或极显著性差异(P<0.05或P<0.01)。结论:氯沙坦能够改善慢性心衰大鼠左心肥厚、左心和血管内皮功能;降低心肌和血管组织中MDA含量并提高SOD活性,减轻组织氧化应激,对心血管产生有益作用。

목적:주요탐토록사탄대5/6신절제만성심쇠모형대서심혈관적영향。방법:웅성SD대서30지수궤분위가수술조(Sham)、병리조(Nx)화록사탄치료조(Lst)。 Nx조화Lst조행5/6신절제술,Lst조우술후제7주초우음수중련속량주급여록사탄(10 mg·kg-1·d-1)치료。8주말처리소유대서,측정혈류동역학삼수、심지수、혈관서장도、혈청기항、뇨소담이급심기화혈관조직중병이철(MDA)함량화초양화물기화매(SOD)활성。결과:여Nx조비교,Lst조대서LVEDP강저,LVSP、LVDP절대치승고,심지수강저,혈관서장도증대,혈청중기항、뇨소담강저,각조직중MDA함량강저,SOD활성승고,결과유현저성차이혹겁현저성차이(P<0.05혹P<0.01)。결론:록사탄능구개선만성심쇠대서좌심비후、좌심화혈관내피공능;강저심기화혈관조직중MDA함량병제고SOD활성,감경조직양화응격,대심혈관산생유익작용。
Objevtives: To explore the cardiovascular effects of losartan in 5/6 nephrectomy rats with chronic heart failure (CHF). Methods: Thirty male SD (Sprague Dawley) rats were randomly divided into sham group(sham), pathology group(Nx) and losartan group(Lst). Nx group and Lst group were induced by 5/6 nephrectomy. Rats in the Lst group were given losatan (10 mg·kg-1·d-1) through drinking water from the beginning of the postoperative 7th week to the end of the 8th week. At the end of the 8th week, all rats were measured for hemodynamic parameters, cardiac index, vasodilation degrees, serum creatinine and urea nitrogen, and the malondialdehyde (MDA) contents and superoxide dismutase (SOD) activities in myocardium and aorta tissues. Results: Compared with the Nx group, rats in the Lst group had decreased LVEDP, cardiac index, serum urea nitrogen and creatinine, and tissue MDA content; and increased LVSP, LVDP, vasodilation and tissue SOD activity. The differences were significant or very significant (P<0.05 or P<0.01). Conclusion: Losartan can improve left ventricular hypertrophy and the left ventricular and vascular en-dothelial functions of rats with CHF. It also may have beneficial effects to the myocardium and aorta tis-sues by reducing tissue oxidative stress of CHF rats.
