该实验采用双水相萃取法提取米糠脂肪酶,通过确定米糠脂肪酶最佳提取条件,为日后大规模生产提供理论依据。用蒸馏水对米糠脂肪酶粗提取实验,通过单因素实验确定最佳提取工艺参数:料液比1∶15,提取温度40℃,提取pH 7.0,提取时间1 h。双水相萃取对粗酶液纯化实验,对影响双水相萃取各种因素进行探讨,并通过正交实验优化提取条件,于PEG浓度22.5%,硫酸铵浓度20%,pH 7.0条件下进行双水相萃取,脂肪酶纯化倍数最高。

This experiment adopts the aqueous two phase extraction method to extract rice bran lipase, through determine the optimal extraction conditions of rice bran lipase,provide theoretical basis for the mass production in the future. With distilled water to the rice bran lipase crude extraction experiment, the best extraction technology was determined by the single factor experiment parameters:the ratio of material to water 1∶15,extraction temperature 40℃,the extraction of pH 7.0,extracting time of 1h. Aqueous two-phase extraction of crude enzyme liquid purification experiment,discusses the factors affecting the aqueous two-phase extraction,and through the orthogonal experiment to optimize the extraction conditions of PEG concentration 22.5%,20% ammonium sulfate concentration,aqueous two-phase extraction under the condition of pH 7.0,lipase purified the highest multiples.
