为鉴定潜在的松材线虫防治靶标基因,应用SL法克隆到松材线虫Rbx1,并完成Rbx1蛋白质结构分析。松材线虫Rbx1与泛素途径多个蛋白发生互作。通过Rbx1与E2的分子对接研究,共发现Rbx1与E2分子间存在3个氢键,Rbx1含有1个E2契合“口袋”。分子动力学模拟表明,Rbx1的氮末端是其功能区域。松材线虫Rbx1是潜在的防治靶标基因,以该区域为靶位点设计抑制子或可达到防治该线虫的目的。
為鑒定潛在的鬆材線蟲防治靶標基因,應用SL法剋隆到鬆材線蟲Rbx1,併完成Rbx1蛋白質結構分析。鬆材線蟲Rbx1與汎素途徑多箇蛋白髮生互作。通過Rbx1與E2的分子對接研究,共髮現Rbx1與E2分子間存在3箇氫鍵,Rbx1含有1箇E2契閤“口袋”。分子動力學模擬錶明,Rbx1的氮末耑是其功能區域。鬆材線蟲Rbx1是潛在的防治靶標基因,以該區域為靶位點設計抑製子或可達到防治該線蟲的目的。

An Rbx1 enzyme gene entire cDNA was cloned from Bursaphelenchus xylohp ilus.The structure and functions of the Rbx1 were analyzed .The Rbx1 and E2 interface was supported by several obvious hydrogen bonding interactions .From the docking of Rbx1 and E2, residues of E2 binding to the central groove provided by the Rbx1.By the molecular dynamics simulation, the N terminus of Rbx1 are important in the binding.The N terminus can be used as the repressor binding site to control the B.xylophilus.Therefore, Rbx1 is a potential target for developing potent antinematodal drugs against B.xy-lophilus.
