目的研究三七叶总皂苷最佳提取工艺及其抗抑郁活性。方法采用正交实验，考察乙醇浓度、提取温度、固液比及提取时间四个因素对三七叶总皂苷提取工艺的影响，利用D101大孔树脂分离，溶剂沉淀纯化三七叶总皂苷。采用小鼠强迫游泳实验和小鼠尾悬挂实验对三七叶总皂苷进行抗抑郁活性评价。结果确立了三七叶总皂苷的最佳提取工艺为：提取时间1行．5 h，乙醇浓度60％，固液比1∶10，提取温度为80℃。三七叶总皂苷与对照品氟西汀类似，均能显著缩短小鼠游泳和悬尾的不动时间。结论优选出的最佳提取工艺条件及纯化总皂苷的方法对进一步的工艺开发有较好的参考价值，所得三七叶总皂苷具有很好的抗抑郁活性。

Objective To study the optimum extracting technology and its antidepressant activity of the total saponins extracted from Sanchi leaf .Methods The orthogonal experiment was adopted to observe the effects of ethanol concentration,extraction temperature,ratio of solid to liquid and extraction time on the total saponins extraction efficiency , and the total saponins of Sanchi leaf were separated and purified by D-101 porous resin absorption and step-sedimentation with acetone-ether.The antidepressant effects of the total saponins of Sanchi leaf were evaluated by forced swimming test and the tail suspension test in mice .Results The optimum conditions of extracting total saponins from Sanchi leaf were extraction time:1.5h,ethanol concentration:60%,solid /lipuid ratio:1∶10 and extraction temperature:80℃.As compared with total saponins from Sanchi leaf like the control drug-fluoxetine could significantly shorten immobility time of swimming and tail suspension in mice .Conclusion The optimum extraction condition and purification method are very useful for further development of industrial process.The total saponins extracted from Sanchi leaf has effective antidepressant activity .
