目的:探讨β-细辛醚对Aβ25-35所致PC12细胞凋亡的抑制作用。方法以Aβ25-35建造体外PC12细胞损伤模型,观察β-细辛醚对其细胞存活率、细胞形态、乳酸脱氢酶( LDH)的含量和Caspase-3、Caspase-6、Caspase-9 mRNA表达的影响。结果10μmol/L的Aβ25-35与PC12细胞共培养24 h出现明显损伤,细胞活力下降,培养液中LDH含量增多,Caspase-3、Caspase-6、Caspase-9 mRNA表达水平增高。15 mg/L、30 mg/L、60 mg/Lβ-细辛醚可有效改善Aβ25-35引起的PC12细胞形态改变,提高细胞存活力,减少LDH渗漏,明显降低Aβ25-35引起的PC12细胞Caspase-3、Caspase-6、Caspase-9 mRNA表达水平。结论β-细辛醚对Aβ25-35所致PC12细胞损伤有保护作用。

목적:탐토β-세신미대Aβ25-35소치PC12세포조망적억제작용。방법이Aβ25-35건조체외PC12세포손상모형,관찰β-세신미대기세포존활솔、세포형태、유산탈경매( LDH)적함량화Caspase-3、Caspase-6、Caspase-9 mRNA표체적영향。결과10μmol/L적Aβ25-35여PC12세포공배양24 h출현명현손상,세포활력하강,배양액중LDH함량증다,Caspase-3、Caspase-6、Caspase-9 mRNA표체수평증고。15 mg/L、30 mg/L、60 mg/Lβ-세신미가유효개선Aβ25-35인기적PC12세포형태개변,제고세포존활력,감소LDH삼루,명현강저Aβ25-35인기적PC12세포Caspase-3、Caspase-6、Caspase-9 mRNA표체수평。결론β-세신미대Aβ25-35소치PC12세포손상유보호작용。
Objective It is to approach the protective effect of β-asarone on PC12 cells apoptosis induced by β-amyloid protein (Aβ25-35).Methods The model of apoptosis of PC12 cells were induced by Aβ25-35 in vitro, the rate of cell viability, the morphological change of PC 12, the consistence of lactate dehydrogenase (LDH), and the expression of Caspase-3, -6, -9 mRNA were observed for assessing the effect on β-asarone in PC12 cells induced by Aβ25-35 .Results Cell survival decrea-sing, LDH leakage and Caspase -3, -6, -9 mRNA expression increasing were happened in PC 12 cells cultured with 10 umol/L pre aggregated Aβ25-35 for 24 hour.15mg/L, 30 mg/L, 60 mg/L β-asarone could increase cell survival , and de-crease the level of lactate dehydrogenase and the expression of Caspase -3, -6, -9 mRNA.Conclusion β-asarone can significantly prevent PC 12 cells apoptosis induced by Aβ25-35 .
