Tanreqing injection%radiation pneumonitis%IL-6%TNF-α%TGF-β1%NF-κB
目的:探讨痰热清注射液防治放射性肺炎的作用及其机制。方法40只雌性SD大鼠随机分为正常组、痰热清组、地塞米松组和模型组各10只,除正常组外其余各组大鼠均用直线加速器对大鼠双肺进行单次照射25 Gy造模。造模后第1天起模型组和正常组予生理盐水灌胃,其余2组分别用痰热清、地塞米松灌胃,连续6周后处死,观察各组大鼠肺系数及肺组织病理改变,检测血清IL-6、TNF-α、TGF-β1的水平和肺组织中 P65核转位水平及磷酸化IκB的表达。结果痰热清组及地塞米松组肺系数、肺组织炎性及纤维化改变、血清IL-6、TNF-α、TGF -β1水平均显著低于模型组(P均<0.01),且上述各指标痰热清组均优于地塞米松组( P均<0.01);痰热清组和地塞米松组细胞核 P65和磷酸化IκB的表达均显著低于模型组(P均<0.01)。结论痰热清注射液可能通过抑制P65的核转位及IκB的磷酸化,抑制血清中IL-6、TNF-α、TGF-β1的合成,从而对大鼠放射性肺炎起到防治作用,反向验证了热毒为放射性肺炎的主要病机之一。

목적:탐토담열청주사액방치방사성폐염적작용급기궤제。방법40지자성SD대서수궤분위정상조、담열청조、지새미송조화모형조각10지,제정상조외기여각조대서균용직선가속기대대서쌍폐진행단차조사25 Gy조모。조모후제1천기모형조화정상조여생리염수관위,기여2조분별용담열청、지새미송관위,련속6주후처사,관찰각조대서폐계수급폐조직병리개변,검측혈청IL-6、TNF-α、TGF-β1적수평화폐조직중 P65핵전위수평급린산화IκB적표체。결과담열청조급지새미송조폐계수、폐조직염성급섬유화개변、혈청IL-6、TNF-α、TGF -β1수평균현저저우모형조(P균<0.01),차상술각지표담열청조균우우지새미송조( P균<0.01);담열청조화지새미송조세포핵 P65화린산화IκB적표체균현저저우모형조(P균<0.01)。결론담열청주사액가능통과억제P65적핵전위급IκB적린산화,억제혈청중IL-6、TNF-α、TGF-β1적합성,종이대대서방사성폐염기도방치작용,반향험증료열독위방사성폐염적주요병궤지일。
Objective It is to approach the effect of Tanreqing injection in inhibit radiation pneumonitis and explore its mechanism .Methods 40 healthy SD rats were divided into normal group , irradiation with Tanreqing group , dexamethasone group and model group, 10 cases in each group.The animals were given corresponding drugs by gavage treatment for 6 weeks. The determination of lung coefficient , pulmonary fibrosis score and pathological changes of lung with Hematoxylin and eosin ( HE) staining were explored to observe the level of radiation pneumonitis .Enzyme-linked immunosorbent assay ( ELISA) was applied to detect the plasma interleukin -6 (IL-6), necrosis factor-α(TNF-α) and transforming growth factor β1 (TGF-β1 ) levels.Western-Blot was applied to detect the translocation of P 65.And in order to investigate the molecular basis of the altered subcellular localization of P 65, phospho-IκBαand total IκBαprotein was tested still by Western-blot method.Re-sults The lung coefficient, pulmonary fibrosis score and the expression of IL -6, TNF-αand TGF-β1 inTanreqing group and dexamethasone group were remarkably lower than that in model group (all P<0.01), and these indexes in Tanreqing group were significantly lower than that in dexamethasone group (all P<0.01).The expression of P65 in nucler and phospho-IκBαin in Tanreqing group and dexamethasone group was remarkable higher than that of the model groups ( all P<0.01 ) . Conclusion Tanreqing injgection can inhibit radiation pneumonitis , whose mechanism probably related to downregulation the expression of P65 in nucler and phospho -IκBα, and represses the expression of IL -6 , TNF-αand TGF-β1 , reverse proved that deficiency of Re and Du is one of traditional Chinese medicine main pathogenesis in radiation pneumonitis .
