目的探讨脂多糖( LPS)促进人工关节松动的可能性。方法采用酶联免疫吸附法(ELISA)测定在不同剂量LPS作用下钛颗粒诱导人外周血单核细胞(MOs)分泌肿瘤坏死因子a(TNF-a)、白细胞介素6(IL-6)、白细胞介素1(IL-1)的含量变化。结果与空白对照组 A 组相比LPS组、钛颗粒组及不同浓度的 LPS联合钛颗粒刺激组刺激后 TNF-a、 IL-6、IL-1即显著升高(均P<0.05), TNF-a、 IL-6、IL-1分泌与LPS呈现浓度依赖性, F组TNF-a、 IL-6、IL-1分泌明显高于A、B、C、D、E组(P<0.05)。结论 LPS能有效地促进由于钛颗粒微粒刺激MOs所致的TNF-a 、IL-6的分泌,增强了人工关节置换术后由微粒诱导的骨溶解。

Objectives To explore the possibility of Lipopolysaccharide (LPS) contribute to aseptic loosening of orthopedic implants. Methods Peripheral blood samples were obtained from healthy human donors. Monocytes (MOs) were isolated and assigned to 6 groups. Group A was treated with MOs alone , group B with MOs + LPS (100ng/ml), group C with MOs+Ti particles, group D,E,F with MOs + Ti particles + LPS (1ng/ ml,10ng/ ml,100ng/ ml), The cel s were co-cultured for 24 hours and the mixtures were tested for osteolytic factors includingTNF-a,IL-6 and IL-1. Results The released contents of TNF-a,IL-6 and IL-1 from MOs were increased significantly compared with those before LPS or Ti particles chal enge (P<0.05).And the content of TNF-a,IL-6 and IL-1 were gradual y increased by LPS With dif erent concentrations (P<0.05) Conclusion The inflammatory cytokine of TNF-a,IL-6 and IL-1 Was increased in the MOs stimulated by LPS.LPS and Ti particles cooperative interact to enhance the biological ef ects of aseptic loosening.
