目的探讨不同冻存温度对树突状细胞存活率的影响，为临床树突状细胞疫苗的贮存提供依据。方法本研究选取健康人的外周血分离单核淋巴细胞，体外诱导培养，成熟的树突状细胞置于-80℃和液氮中的温度下贮存。不同的时间复苏，苔盼蓝染色计算树突状细胞的存活率。结果在一周之内树突状细胞在-80℃和液氮中冻存复苏后，活细胞数85%以上，四周后在液氮中存活的树突状细胞的数量明显高于-80℃冻存的细胞。结论液氮中适合长期保存树突状细胞。

Objective: explore to the effects of frozen temperature on the survival of dendritic cel s (DCs), and provide the basis for the storage of clinical DCs vaccine. Methods:Peripheral blood mononuclear cel s were isolated from healthy people, and induced into DCs in vitro. The mature DCs were stored in -80 ℃ and liquid nitrogen temperature. The survival of DCs was analysed by typan blue staining after thawed in different intervals. Results: The survivals of DCs thawed within a week was more than 85% both in -80℃ and liquid nitrogen cryopreservation. However the survivals of DCs in liquid nitrogen cryopreservation was obviously higher than that in -80℃ thawed in four weeks later. Conclusion: DCs is more optimal for long-term preservation in liquid nitrogen than in -80℃.
