目的探讨整合素链接激酶(integrin-linked kinase,ILK)在非小细胞肺癌(non-small cell lung cancer,NSCLC)的表达情况及与临床病理因素、微血管密度( microvessel density, MVD)之间的关系。方法对101例NSCLC及相应癌旁正常肺组织行抗ILK免疫组织化学( SP法)染色;对101例NSCLC行抗CD34免疫组织化学染色并进行MVD计数。结果 ILK在正常支气管上皮细胞中为阴性表达,而在NSCLC组织中出现胞质或胞质/胞核同时表达,其总的阳性率为65.35%(66/101),与正常肺组织比较差异具有显著性(P<0.001)。无淋巴结转移组中ILK阳性率[51.16%(22/43)]显著低于淋巴结转移组[75.86%(44/58),P=0.010]。Ⅰ、Ⅱ期NSCLC患者ILK阳性率[52.27%(23/44)]显著低于其在Ⅲ、Ⅳ期患者的阳性率[75.44%(43/57),P=0.015]。 NSCLC中,ILK阳性表达组的MVD值显著高于ILK阴性表达组(47.02±7.39 vs.37.11±8.40,t=6.109,P<0.001)。结论 NSCLC组织中ILK表达显著高于正常肺组织,并与NSLCL的淋巴结转移,TNM分期及MVD相关,提示ILK可能通过促进血管形成而促进NSCLC的进展。

목적탐토정합소련접격매(integrin-linked kinase,ILK)재비소세포폐암(non-small cell lung cancer,NSCLC)적표체정황급여림상병리인소、미혈관밀도( microvessel density, MVD)지간적관계。방법대101례NSCLC급상응암방정상폐조직행항ILK면역조직화학( SP법)염색;대101례NSCLC행항CD34면역조직화학염색병진행MVD계수。결과 ILK재정상지기관상피세포중위음성표체,이재NSCLC조직중출현포질혹포질/포핵동시표체,기총적양성솔위65.35%(66/101),여정상폐조직비교차이구유현저성(P<0.001)。무림파결전이조중ILK양성솔[51.16%(22/43)]현저저우림파결전이조[75.86%(44/58),P=0.010]。Ⅰ、Ⅱ기NSCLC환자ILK양성솔[52.27%(23/44)]현저저우기재Ⅲ、Ⅳ기환자적양성솔[75.44%(43/57),P=0.015]。 NSCLC중,ILK양성표체조적MVD치현저고우ILK음성표체조(47.02±7.39 vs.37.11±8.40,t=6.109,P<0.001)。결론 NSCLC조직중ILK표체현저고우정상폐조직,병여NSLCL적림파결전이,TNM분기급MVD상관,제시ILK가능통과촉진혈관형성이촉진NSCLC적진전。
Objective To investigate the expression of integrin-linked kinase ( ILK) and its correlation with clinicopathological parameters and microvessel density ( MVD ) in non-small cell lung cancer ( NSCLC ) . Methods Immunohistochemical method were used to analyze the expression of ILK in 101 cases of NSCLC and adjacent non-neoplasitc lung tissues , and MVD was analyzed by CD 34 immunostaining .Results The immunohistochemistry results showed that the positive expression rate of ILK in NSCLC was 65.35%(66/101),but the expression of ILK was not detected in normal bronchial epithelium ( P<0.001 ) .The positive ILK expression was directly associated with advanced TNM stage ( P =0.015 ) , and lymph metastasis ( P =0.010 ) .There was no correlation between the expression of ILK and other clinicopathological factors ( P>0.05 ) .The MVD in positive ILK-staining cases was much higher than that in negative ILK-staining cases (47.02 ±7.39 vs.37.11 ±8.40,t=6.109, P<0.001).Conclusions The expression of ILK is significantly up-regulated in NSCLC,and positively correlated with lymphatic metastasis,advanced TNM stage,and MVD.ILK may be involved in angiogenesis and thus enhance the progression of NSCLC .
