目的探讨姜黄素对脊髓缺血损伤后脂质过氧化诱发细胞凋亡的影响。方法建立新西兰大白兔脊髓缺血模型,分为缺血对照组、姜黄素给药组及假手术组。采用神经功能评分评价脊髓缺血兔后肢运动功能。采用硫代巴比妥酸(TBA )比色法、黄嘌呤氧化酶法、硝酸还原酶法、RT-PCR和流式细胞仪等技术检测丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表达水平和细胞凋亡率。结果与缺血对照组比较,姜黄素可降低MDA含量、NO含量和Caspase-3表达量,提高SOD活性,降低细胞凋亡率,差别有统计学意义。神经功能评分结果显示姜黄素给药组评分显著高于缺血对照组。结论姜黄素对兔脊髓缺血损伤具有保护作用,此作用与抑制脂质过氧化、从而减少缺血细胞凋亡有关。

목적탐토강황소대척수결혈손상후지질과양화유발세포조망적영향。방법건립신서란대백토척수결혈모형,분위결혈대조조、강황소급약조급가수술조。채용신경공능평분평개척수결혈토후지운동공능。채용류대파비타산(TBA )비색법、황표령양화매법、초산환원매법、RT-PCR화류식세포의등기술검측병이철(MDA)、초양화물기화매(SOD)、일양화담(NO)、반광안산천동안산단백매3(Caspase-3)적표체수평화세포조망솔。결과여결혈대조조비교,강황소가강저MDA함량、NO함량화Caspase-3표체량,제고SOD활성,강저세포조망솔,차별유통계학의의。신경공능평분결과현시강황소급약조평분현저고우결혈대조조。결론강황소대토척수결혈손상구유보호작용,차작용여억제지질과양화、종이감소결혈세포조망유관。
Objective To observed the effects of curcumin on apoptosis induced by lipid peroxida-tion after spinal cord ischemia . Methods Spinal cord ischemia model of New Zealand white rabbits were established ,and then divided into ischemic control group ,curcumin dose group and the sham-operation group randomly . Apply neurological score to evaluate the spinal cord ischemic rabbit hindlimb function . Apply thiobarbituric acid ,xanthine oxidase ,nitrate reductase method ,RT-PCR and flow cytometry to detect the concentration of malondialdehyde(MDA) ,superoxide dismutase(SOD) ,nitric oxide(NO) ,the expression levels of caspase 3 and apoptosis rate . Results Compared to ischemic control group ,curcu-min can reduce the concentration of MDA ,NO and Caspase-3 expression levels ,increase SOD activity , reduce the rate of apoptosis ,the difference was statistically significant . The neurological score results show that curcumin treatment group scores were significantly higher than the ischemic control group . Conclusion This study showed that curcumin has a protective effect on spinal cord ischemic injury ,which may be related to its inhibition of lipid peroxidation ,thereby reducing the ischemic cell apoptosis .
