目的：探讨ATR基因的表达对HeLa细胞对烷化剂敏感性的影响。方法：采用RNA干扰技术抑制HeLa细胞ATR基因的表达，观察ATR被阻断后HeLa细胞对烷化剂敏感性的变化，从而确定ATR基因的作用。结果：筛选到表达针对ATR基因的短发夹RNA（shRNA）阳性克隆，Western印迹结果显示ATR基因表达受到明显抑制；HeLa细胞对烷化剂的敏感性实验提示，ATR shRNA+ HeLa细胞对烷化剂的敏感性明显增强。结论：抑制ATR基因可明显提高HeLa细胞对烷化剂的敏感性。
目的：探討ATR基因的錶達對HeLa細胞對烷化劑敏感性的影響。方法：採用RNA榦擾技術抑製HeLa細胞ATR基因的錶達，觀察ATR被阻斷後HeLa細胞對烷化劑敏感性的變化，從而確定ATR基因的作用。結果：篩選到錶達針對ATR基因的短髮夾RNA（shRNA）暘性剋隆，Western印跡結果顯示ATR基因錶達受到明顯抑製；HeLa細胞對烷化劑的敏感性實驗提示，ATR shRNA+ HeLa細胞對烷化劑的敏感性明顯增彊。結論：抑製ATR基因可明顯提高HeLa細胞對烷化劑的敏感性。
목적：탐토ATR기인적표체대HeLa세포대완화제민감성적영향。방법：채용RNA간우기술억제HeLa세포ATR기인적표체，관찰ATR피조단후HeLa세포대완화제민감성적변화，종이학정ATR기인적작용。결과：사선도표체침대ATR기인적단발협RNA（shRNA）양성극륭，Western인적결과현시ATR기인표체수도명현억제；HeLa세포대완화제적민감성실험제시，ATR shRNA+ HeLa세포대완화제적민감성명현증강。결론：억제ATR기인가명현제고HeLa세포대완화제적민감성。
Objective: To investigate the effect of the expression of ATR gene on HeLa cells' susceptibility to al-kylating agents. Methods: RNA interference technology was employed to suppress the expression of ATR, then the role of ATR in HeLa cells' anti-alkylation ability was inspected. Results: A positive clone of siRNA against ATR gene was successfully screened and it can strikingly inhibit the expression of ATR in HeLa cells by Western blot analysis, and then the ability of HeLa cells to resist alkylating agents was significantly attenuated. Conclusion:The inhibition of the expression of ATR gene can significantly improve the HeLa cells' susceptibility to alkylating agents.
