以拟南芥油菜素内酯受体BRI1基因序列设计同源简并引物,利用同源克隆技术,克隆了甘蓝型油菜中编码油菜素内酯受体基因全长 cDNA 序列,命名为BnBRI1(GenBank:JX871217),该基因开放阅读框(ORF)大小为3591 bp,与拟南芥中BRI1基因(GenBank:NM_120100)相似性为94.21%,编码1196个氨基酸,与拟南芥中BRI 编码的氨基酸相似性为89.23%.采用实时定量PCR分析表明,BnBRI1基因在根、茎、叶中都有表达,但表达量存在显著差异.在两叶一心期和花期的根中表达量最高,四叶一心期和抽薹期的叶中表达量最高.

Homologous degenerented primers were designed based on the sequence of the brassinolide re-ceptor gene BRI1 in Arabidopsis thaliana.The method of homologous clone was used to clone and ana-lyze a full-length cDNA of the brassinolide receptor gene in Brassica napus L.which was named Bn-BRI1 (GenBank:JX871217 ).The 3591bp-length of the whole sequence of the open reading frame (ORF)of BnBRI had 94.21% similarity with Arabidopsisthaliana BRI (GenBank:NM_120100).For the putative protein,the protein of BnBRI1 contained 1196 amino acids having 89.23%similarity with the BRI1 protein inArabidopsis thaliana.Then the expression of BnBRI1 in different tissues of the dif-ferent development stages in B.napus L.was analyzed using real-time polymerase chain reaction.This result showed that the expression ofBnBRI1 existed in different tissues of the different development pe-riods,but the amount of expression varied,highest in the root of two-leaf and flower period and in the leaf of four-leaf period and bolting period.
