目的建立黄芪桂枝五物汤中的黄芪甲苷的含量测定方法。方法采用HPLC-ELSD法测定黄芪桂枝五物汤中黄芪甲苷的含量,色谱条件为:Welchrom-C18色谱柱(250 mm ×4.6 mm ,5μm),以乙腈-水(32∶68)为流动相,流速为1 mL · min-1,柱温25℃;ELSD参数:漂移管温度75℃,喷雾管温度40℃,气体流量1.5 L · min-1,输出压力0.5 M Pa。结果黄芪甲苷在0.03~1.22 mg · mL -1范围内呈良好的线性关系(r=0.9995),平均回收率为99.8%,方法精密度的RSD为2.1%(n=6)。结论该方法简单、准确、重复性好,可用于黄芪桂枝五物汤的质量控制。

목적건립황기계지오물탕중적황기갑감적함량측정방법。방법채용HPLC-ELSD법측정황기계지오물탕중황기갑감적함량,색보조건위:Welchrom-C18색보주(250 mm ×4.6 mm ,5μm),이을정-수(32∶68)위류동상,류속위1 mL · min-1,주온25℃;ELSD삼수:표이관온도75℃,분무관온도40℃,기체류량1.5 L · min-1,수출압력0.5 M Pa。결과황기갑감재0.03~1.22 mg · mL -1범위내정량호적선성관계(r=0.9995),평균회수솔위99.8%,방법정밀도적RSD위2.1%(n=6)。결론해방법간단、준학、중복성호,가용우황기계지오물탕적질량공제。
Objective To establish an HPLC-ELSD method for determination of astragaloside Ⅳ in Huangqi Guizhi Wuwu Decoc-tion .Methods The astragaloside Ⅳ in the decoction was determined by HPLC-ELSD .The experiment conditions were as fol-lows:Welchrom-C18 (250 mm × 4 .6 mm ,5 μm) column and a mixture of acetonitrile-water (32∶68) as mobile phase .The flow rate was 1 .0 mL · min-1 and the temperature of drift tube for measurement was 75 ℃ .The pressure of air was 0 .5 MPa and the flow rate of carrier gas was 1 .5 L · min-1 .Results There was a good linearity within the range of 0 .03-1 .22 mg · mL -1 (r=0 .999 5) for astragaloside Ⅳ ,and the average recovery was 99 .8% with RSD of 2 .1% .Conclusion The method is simple ,accu-rate and reproducible .It can be used as the quality control method of Huangqi Guizhi Wuwu Decoction .
