目的检测胰岛素样生长因子Ⅰ( IGF-Ⅰ)、转化生长因子β2( TGF-β2)单独及联合应用对立体培养软骨细胞增殖速度、细胞形态和表型、黏多糖含量及Ⅱ型胶原含量的影响。方法取兔第二代软骨细胞于藻酸钙凝珠中,分别加入IGF-Ⅰ、TGF-β2及IGF-Ⅰ+TGF F-β2进行立体培养。 HE、甲苯胺蓝染色测软骨细胞形态、胞外基质变化;阿尔新蓝法测黏多糖;免疫组化法测Ⅱ型胶原及表型;Woessner法测Ⅱ型胶原含量。结果细胞形态及表型的维持、增殖速度、黏多糖及Ⅱ型胶原含量, IGF-Ⅰ+TGF-β2组明显优于其他组,各组间差异均具有统计学意义( P<0.05)。结论 IGF-Ⅰ与TGF-β2联合应用起协同作用,能够显著促进黏多糖及Ⅱ型胶原的分泌,促进软骨细胞外基质分泌及软骨细胞形态及表型的维持。

목적검측이도소양생장인자Ⅰ( IGF-Ⅰ)、전화생장인자β2( TGF-β2)단독급연합응용대입체배양연골세포증식속도、세포형태화표형、점다당함량급Ⅱ형효원함량적영향。방법취토제이대연골세포우조산개응주중,분별가입IGF-Ⅰ、TGF-β2급IGF-Ⅰ+TGF F-β2진행입체배양。 HE、갑분알람염색측연골세포형태、포외기질변화;아이신람법측점다당;면역조화법측Ⅱ형효원급표형;Woessner법측Ⅱ형효원함량。결과세포형태급표형적유지、증식속도、점다당급Ⅱ형효원함량, IGF-Ⅰ+TGF-β2조명현우우기타조,각조간차이균구유통계학의의( P<0.05)。결론 IGF-Ⅰ여TGF-β2연합응용기협동작용,능구현저촉진점다당급Ⅱ형효원적분비,촉진연골세포외기질분비급연골세포형태급표형적유지。
Objective To investigate the effects of insulin-like growth factorⅠ( IGF-Ⅰ) , transformation growth fator beta2(TGF-β2)alone and combined application on proliferation rate ,cell morphology,glycosaminoglycan content ,typeⅡcollagen content and cell phenotype of chondrocytes for three -dementional culture .Methods The second generation chondrocytes of rabbit were seeded in calcium alginate beads ,and IGF-Ⅰ,TGF-β2 and IGF-Ⅰ+TGF-β2 were added respectively for three-dementional culture .HE staining and toluidine blue staining were used to measure chondrocytes morphology and extracellular matrix change .Alcian blue mothod was used to measure mucopolysaccharides content .Immunohistochemical method was applied to detect typeⅡcollagen and cell phenotype . Results Maintainence of cell morphology and phenotype ,proliferation rate ,mucopolysaccharides and typeⅡcollagen of chondrocytes in IGF-Ⅰ +TGF-β2 group were significantly higher than IGF-Ⅰ or TGF-β2 alone. Conclusions Combination of IGF-Ⅰand TGF-β2 could promote extracellular matrix secretion of chondrocytes and maintainence of cell morphology and phenotype ,as well as mucopolysaccharides secretion .
