目的探讨骨髓间充质干细胞(MSCs)在溃疡性结肠炎(UC)大鼠发病中的作用,并分析可能与调节Treg及Th17细胞有关。方法用三硝基苯磺酸( TNBS )诱导建立UC大鼠模型,体外培养扩增雄性SD大鼠MSCs,建立空白对照组,未作任何处理;模型组,仅用TNBS灌肠(3 ml/kg);MSCsⅠ组,MSCsⅡ组和MSCsⅢ组分别用TNBS(3 ml/kg),3 d后,MSCsⅠ组给予尾静脉注射低剂量MSCs(1×105),MSCsⅡ组给予尾静脉注射中剂量MSCs(1×106),MSCsⅢ组给予尾静脉注射高剂量MSCs(1×107)。计算各组大鼠的疾病活动指数( DAI)、大体形态评分( GMS)和结肠组织学病理评分( HDS),并通过免疫组化、流式细胞术观察MSCs对UC大鼠Treg/Th17的影响。结果 DAI、GMS和HDS在模型组中高于空白对照组,Treg细胞在UC大鼠模型中表达下调,Th17细胞在模型组中表达上调,差异有统计学意义( P<0.05)。与模型组相比,MSCs治疗组DAI、GMS和HDS降低,且MSCsⅡ组低于MSCsⅠ组与MSCsⅢ组,结肠组织与外周血Treg细胞表达上调, MSCsⅡ组高于MSCsⅠ组与MSCsⅢ组,差异有统计学意义( F=9.25,641.00,P<0.05);Th17细胞表达下降, MSCsⅡ组低于MSCsⅠ组与MSCsⅢ组,差异有统计学意义( F=15.95,318.80,P<0.05)。结论 MSCs通过对UC大鼠Treg、Th17的影响,对UC大鼠起到治疗作用。

목적탐토골수간충질간세포(MSCs)재궤양성결장염(UC)대서발병중적작용,병분석가능여조절Treg급Th17세포유관。방법용삼초기분광산( TNBS )유도건립UC대서모형,체외배양확증웅성SD대서MSCs,건립공백대조조,미작임하처리;모형조,부용TNBS관장(3 ml/kg);MSCsⅠ조,MSCsⅡ조화MSCsⅢ조분별용TNBS(3 ml/kg),3 d후,MSCsⅠ조급여미정맥주사저제량MSCs(1×105),MSCsⅡ조급여미정맥주사중제량MSCs(1×106),MSCsⅢ조급여미정맥주사고제량MSCs(1×107)。계산각조대서적질병활동지수( DAI)、대체형태평분( GMS)화결장조직학병리평분( HDS),병통과면역조화、류식세포술관찰MSCs대UC대서Treg/Th17적영향。결과 DAI、GMS화HDS재모형조중고우공백대조조,Treg세포재UC대서모형중표체하조,Th17세포재모형조중표체상조,차이유통계학의의( P<0.05)。여모형조상비,MSCs치료조DAI、GMS화HDS강저,차MSCsⅡ조저우MSCsⅠ조여MSCsⅢ조,결장조직여외주혈Treg세포표체상조, MSCsⅡ조고우MSCsⅠ조여MSCsⅢ조,차이유통계학의의( F=9.25,641.00,P<0.05);Th17세포표체하강, MSCsⅡ조저우MSCsⅠ조여MSCsⅢ조,차이유통계학의의( F=15.95,318.80,P<0.05)。결론 MSCs통과대UC대서Treg、Th17적영향,대UC대서기도치료작용。
Objective To evaluate the effect of MSCs on rat with ulcerative colitis ,which is related to the regulation of Treg and Th17.Methods We established animal model of ulcerative colitis rats by trinitrobenzene sulphonic acid(TNBS)enema,MSCs from male rat was cultured and amplified in vitro.And we made control group without doing anything ,model group are only clystered by TNBS ,MSCsⅠgroup,MSCsⅡgroup and MSCsⅢgroup are clystered by TNBS,and three days later,MSCsⅠgroup administrated with low dose of MSCs by tail vein (1 ×105);MSCsⅡgroup administrated with medium dose of MSCs by tail vein (1 ×106 );MSCsⅢgroup administrated with high dose of MSCs by tail vein(1 ×107).Then we counted the DAI,GMS and HDS,and detected the influence of Treg and Th17 which is regulated by MSCs on rat with ulcerative colitis by immunohistochemistry and flow cytometry . Results The DAI,GMS,HDS were higher in the model group compared with control group ,Treg was significantly lower in the model group,and Th17 was significantly higher in the model group (P<0.05).The DAI,GMS,HDS in MSCs group were lower than it in model group ,and it was the lowest in the MSCs groupⅡ.Treg was increased ,and it was highest in MSCs groupⅡ( F=9.25 ,641.00 ,P<0.05 ) .Th17 was decreased ,and it was lowest in MSCs groupⅡ( F=15.95 ,318.80 ,P<0.05 ) .Conclusion MSCs plays a therapeutic role on rat with ulcerative colitis via the affection of Treg and Th17.
