目的本研究旨在建立miR-181 b慢病毒载体感染U87胶质瘤干细胞模型后,检测其对TMZ化疗耐受性的变化,并探讨miR-181 b对于恶性脑胶质瘤( GBM )辅助化学治疗的意义。方法通过miR-181 b慢病毒表达载体转染U87胶质瘤干细胞,提高miR-181 b在U87胶质瘤干细胞中的表达。进一步通过RT-PCR、二次神经球形成实验、琼脂糖集落形成实验、MTT实验来分析miR-181 b对于 U87胶质瘤干细胞在TMZ化疗耐受性方面的作用。结果 miR-181 b慢病毒表达载体转染U87胶质瘤干细胞后,U87胶质瘤干细胞中的miR-181b明显上调,而且U87胶质瘤干细胞的生长受到明显抑制;miR-181b可以加强TMZ抑制U87胶质瘤干细胞的生长的作用。结论 miR-181b可以加强TMZ抑制U87胶质瘤干细胞的生长,miR-181b可能对于提高U87胶质瘤干细胞对TMZ的敏感性,降低化疗耐受性起到一定作用。

목적본연구지재건립miR-181 b만병독재체감염U87효질류간세포모형후,검측기대TMZ화료내수성적변화,병탐토miR-181 b대우악성뇌효질류( GBM )보조화학치료적의의。방법통과miR-181 b만병독표체재체전염U87효질류간세포,제고miR-181 b재U87효질류간세포중적표체。진일보통과RT-PCR、이차신경구형성실험、경지당집락형성실험、MTT실험래분석miR-181 b대우 U87효질류간세포재TMZ화료내수성방면적작용。결과 miR-181 b만병독표체재체전염U87효질류간세포후,U87효질류간세포중적miR-181b명현상조,이차U87효질류간세포적생장수도명현억제;miR-181b가이가강TMZ억제U87효질류간세포적생장적작용。결론 miR-181b가이가강TMZ억제U87효질류간세포적생장,miR-181b가능대우제고U87효질류간세포대TMZ적민감성,강저화료내수성기도일정작용。
Objective To establish the miR-181 b lentiviral vector infected the U 87 glioma stem cells model,than detect the U87 glioma stem cells′chemotherapy tolerance to temozolomide (TMZ),and to explore the significance of the miR-181b for malignant glioma (gliomablastoma multiforme,GBM) in the adjuvant chemotherapy. Methods Using the miR-181 b lentiviral vector transfected the U 87 glioma stem cells and improve the level of miR-181b in the U87 glioma stem cells.RT-PCR,Secondary neurospheres formed assay ,agarose colony formation assay and MTT assay were used to detect the role of the miR-181b affect the U87 glioma stem cells in the TMZ chemotherapy tolerance .Results The level of miR-181b in the U87 glioma stem cells was significantly increased after miR-181b lentiviral vectors were transfected the U87 glioma stem cells,and the growth of U87 glioma stem cells were significantly inhibited.Furthermore,miR-181b can strengthen the role of TMZ in the growth inhibition of U87 glioma stem cells .Conclusions We found that miR-181 b can strengthen the role of TMZ in the growth inhibition of U87 glioma stem cells,miR-181b may play a certain role of improving the sensitivity of U 87 glioma stem cells to TMZ and reducing the chemotherapy tolerance .
