目的研究缺氧诱导因子-1α( HIF-1α)和肿瘤细胞增殖抗原Ki67在乳腺癌及癌旁组织及良性肿瘤组织中的表达情况;分析HIF-1α和Ki67之间的相关性,初步探讨HIF-1α对乳腺癌细胞增殖的影响。方法取2008至2010年在桂林医学院附属医院手术治疗后存档并留有病理标本的70例乳腺癌、10例乳腺良性肿瘤组织标本,采用快捷免疫组化S-P法检测乳腺癌组织及其癌旁组织和良性肿瘤组织中HIF-1α和Ki67的表达情况,研究它们与临床病理特征的关系,进一步探讨缺氧对乳腺癌细胞增殖的影响。结果70例乳腺癌组织中HIF-1α和Ki67的阳性表达率分别为78.6%(55/70)和88.6%(62/70),其癌旁组织中HIF-1α和Ki67的阳性表达率分别为45.7%(32/70)和11.4%(8/70),10例乳腺良性肿瘤组织中HIF-1α的阳性表达率为0(0/10),Ki67的表达率均为阴性,差异有统计学意义(P<0.01);其表达与临床分期及腋淋巴结转移密切相关( P<0.05),但与患者年龄、绝经状况、肿块大小、激素受体无明显相关,在乳腺癌组织中HIF-1α与Ki67的表达强度呈正相关( P<0.05),在癌旁组织及良性肿瘤组织未见明显相关( P>0.05)。结论乳腺癌组织中HIF-1α高表达,与淋巴结转移及组织恶性程度相关,提示HIF-1α可能成为预测乳腺癌侵袭、转移和预后的重要指标;HIF-1α和Ki67二者表达强度呈明显正相关,提示缺氧可促进乳腺癌细胞增殖,但确切作用机制有待进一步研究。

Objective To study the expression of hypoxia inducible factor-1α( HIF-1α) and proliferation antigen Ki67 in breast carcinoma ,carcinoma adjacent tissues and benign tumors;the correlation between HIF-1αand Ki67 ,and the role of HIF-1αin the proliferation of breast carcinoma cells .Methods Immunohistochemistry SP method detected the expression of HIF-1αand Ki67 in 70 cases of breast carcinoma sample and adjacent normal tissues,and 10 cases of benign breast tumor tissues ,of whom underwent breast tumor resection in our hospital from 2008 to 2010 , study the role of HIF-1αin the proliferation of breast carcinoma cells .Results The positive expression rates of HIF-1αand Ki67 in 70 cases of breast carcinoma were 78.6%( 55/70 ) and 88.6%( 62/70 ) respectively,and that in adjacent normal tissues were 45.7%(32/70)and 11.4%(8/70).As well as,the positive expression rates of HIF-1αin 10 cases of benign breast tumor tissue was 0(0/10)while that of Ki67 was negative. Therefore,the positive expression rate of HIF-1αand Ki67 in breast cancer was significantly higher than that in adjacent normal tissues and benign breast tumor tissues ( P<0.01 );The positive expression rates of HIF-1αand Ki67 had no significant correlation with age,menopausal status,tumor size and ER.but closely related with lymph node metastasis and clinical stage (P<0.05);The expression intensity of HIF-1αwas correlated with Ki67 in breast carcinoma(P<0.05),but no correlation was indicated in adjacent normal tissues and benign breast tumor tissues (P>0.05 ) .Conclusions The expressions of HIF-1αand Ki67 are high in breast carcinoma ,which are associated with malignancy and lymph node metastasis .There was a positive correlation in the expression intensity of HIF-1αand Ki67 .It suggested that HIF-1αand Ki67 may be affect the biological behavior of breast carcinoma through the interaction,the both may be important predictors for the invasion ,metastasis and prognosis of breast carcinoma .
