[目的]探讨高良姜总黄酮(Total flavonoids from Alpinia officinarum Hance,TFAO)对铅中毒小鼠脑、肾脏氧化应激的保护作用。[方法]60只小鼠分为正常组,模型组,阳性组,TFAO低、中、高剂量组。通过腹腔注射醋酸铅建立铅中毒模型,灌胃给予受试物。实验结束后,测定小鼠体重、脏器指数,血液及组织铅浓度,脑、肾氧化应激参数,如还原型谷胱甘肽(glutathione,GSH)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)等指标。[结果]高浓度TFAO对小鼠血液及组织中铅含量有明显降低作用,且未造成其他必需元素如锌的流失。与模型组相比较,TFAO可显著增加小鼠脑、肾GSH含量和抗氧化酶活性,并能显著降低MDA水平。[结论] TFAO可通过提高铅中毒小鼠组织中抗氧化酶的活性,改善脂质过氧化,从而发挥其对铅诱导的脑、肾氧化应激的保护效应。
[目的]探討高良薑總黃酮(Total flavonoids from Alpinia officinarum Hance,TFAO)對鉛中毒小鼠腦、腎髒氧化應激的保護作用。[方法]60隻小鼠分為正常組,模型組,暘性組,TFAO低、中、高劑量組。通過腹腔註射醋痠鉛建立鉛中毒模型,灌胃給予受試物。實驗結束後,測定小鼠體重、髒器指數,血液及組織鉛濃度,腦、腎氧化應激參數,如還原型穀胱甘肽(glutathione,GSH)、丙二醛(malondialdehyde,MDA)、穀胱甘肽過氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、過氧化氫酶(catalase,CAT)等指標。[結果]高濃度TFAO對小鼠血液及組織中鉛含量有明顯降低作用,且未造成其他必需元素如鋅的流失。與模型組相比較,TFAO可顯著增加小鼠腦、腎GSH含量和抗氧化酶活性,併能顯著降低MDA水平。[結論] TFAO可通過提高鉛中毒小鼠組織中抗氧化酶的活性,改善脂質過氧化,從而髮揮其對鉛誘導的腦、腎氧化應激的保護效應。
[목적]탐토고량강총황동(Total flavonoids from Alpinia officinarum Hance,TFAO)대연중독소서뇌、신장양화응격적보호작용。[방법]60지소서분위정상조,모형조,양성조,TFAO저、중、고제량조。통과복강주사작산연건립연중독모형,관위급여수시물。실험결속후,측정소서체중、장기지수,혈액급조직연농도,뇌、신양화응격삼수,여환원형곡광감태(glutathione,GSH)、병이철(malondialdehyde,MDA)、곡광감태과양화물매(glutathione peroxidase,GSH-Px)、초양화물기화매(superoxide dismutase,SOD)、과양화경매(catalase,CAT)등지표。[결과]고농도TFAO대소서혈액급조직중연함량유명현강저작용,차미조성기타필수원소여자적류실。여모형조상비교,TFAO가현저증가소서뇌、신GSH함량화항양화매활성,병능현저강저MDA수평。[결론] TFAO가통과제고연중독소서조직중항양화매적활성,개선지질과양화,종이발휘기대연유도적뇌、신양화응격적보호효응。
[Objective] To investigate the protective effect of total flavonoids from Alpinia officinarum Hance(TFAO) on lead acetate induced oxidative stress in brain and kidney of mice. [Methods] Al 60 mice were randomly divided into 6 groups:normal control group, lead group, positive control group, lead+TFAO(100, 300, 500 mg·kg-1) groups. Mice except normal control group received intraperitoneal injections of lead acetate every other day, and dif-ferent test substances were administrated to mice oral y once a day. After the last dose was administrated, the body weight, viscera index, lead and zinc con-tents in blood, brain and kidney, oxidative stress parameters such as glutathione(GSH), glutathione peroxidase(GSH-Px), superoxide dismutase(SOD), cata-lase(CAT) and malondialdehyde (MDA) in brain and kidney were determined. [Results] TFAO in high dose (500 mg·kg-1 could lower the lead level in blood, brain, and kidney significantly. The other trace elements such as zinc had no significant decrease after TFAO treatment in blood, however, TFAO in high doses could increase the zinc content in brain, and kidney significantly. Three different doses of TFAO(100, 300, 500 mg·kg-1) could increase the activity of antioxidant enzymes and the content of GSH, decrease the content of MDA in brain and kidney significantly. [Conclusion] TFAO might im-prove antioxidant defense system, reverse lipid peroxidation and protect brain and kidney against lead induced oxidative stress in mice significantly.
