目的通过HE、TUNEL染色镜下观察脊髓损伤,探讨比格犬脊柱图像引导放射治疗( image guided radiation therapy ,IGRT)的可耐受剂量的研究。方法选用6只雄性成年比格犬按放疗计划(单次剂量10Gy、剂量率4Gy/min、5野及射线等角度)对其C7~12椎体及其附件进行 IGRT,放疗2次/w,总剂量依次为50Gy、60Gy、70Gy、80Gy、90Gy、100Gy,并于放疗结束后3个月时活杀取材,取各犬相同节段、相同部位(C7~12)脊髓,采用HE和TUNEL染色光镜下观察脊髓神经细胞形态学的改变。结果比格犬脊髓HE染色结果示:总剂量在80Gy以下的镜下可见轻度充血、水肿,部分神经元失去极性外观变为圆形,树突、轴突消失,尼氏小体淡染,细胞核水肿,偏位;总剂量在80Gy以上时镜下多见凋亡细胞,大量部分神经元失去极性外观,尼氏小体崩解、消失,胞核固缩、碎裂、溶解,出现凋亡小体。比格犬脊髓TUNEL染色示:总剂量在80Gy以下的可见少量凋亡神经细胞,主要集中于脊髓灰质前角和白质;总剂量在80Gy以上的可见较多凋亡神经细胞,主要集中于脊髓灰质前角及白质。结论正常比格犬脊柱IGRT可耐受剂量上限约80Gy左右。

목적통과HE、TUNEL염색경하관찰척수손상,탐토비격견척주도상인도방사치료( image guided radiation therapy ,IGRT)적가내수제량적연구。방법선용6지웅성성년비격견안방료계화(단차제량10Gy、제량솔4Gy/min、5야급사선등각도)대기C7~12추체급기부건진행 IGRT,방료2차/w,총제량의차위50Gy、60Gy、70Gy、80Gy、90Gy、100Gy,병우방료결속후3개월시활살취재,취각견상동절단、상동부위(C7~12)척수,채용HE화TUNEL염색광경하관찰척수신경세포형태학적개변。결과비격견척수HE염색결과시:총제량재80Gy이하적경하가견경도충혈、수종,부분신경원실거겁성외관변위원형,수돌、축돌소실,니씨소체담염,세포핵수종,편위;총제량재80Gy이상시경하다견조망세포,대량부분신경원실거겁성외관,니씨소체붕해、소실,포핵고축、쇄렬、용해,출현조망소체。비격견척수TUNEL염색시:총제량재80Gy이하적가견소량조망신경세포,주요집중우척수회질전각화백질;총제량재80Gy이상적가견교다조망신경세포,주요집중우척수회질전각급백질。결론정상비격견척주IGRT가내수제량상한약80Gy좌우。
Objective Through HE, TUNEL staining of spinal cord injury, explore the Beagle spinal image guided radiotherapy (IGRT) on the tolerance dose.Methods a total of 6 adult male beagle dogs by radiotherapy (10Gy as a single dose, dose rate 4Gy/min, 5 wild and equal angle) of IGRT on the C7~12 vertebral body and its accessories,2 treatments per week, the total dose was 50Gy, 60Gy, 70Gy, 80Gy, 90Gy, 100Gy, and in 3 months after radiotherapy of live animals were put to death,Take the dog of the same segment, the same location (C7~12) spinal cord, HE and TUNEL staining was used to observe the changes of morphology of spinal cord nerve cel s under light microscope .Results Results of the Beagle spinal HE staining:Under the total dose 80Gy,it showed mild hyperemia, edema, neuron loss of polarity appearance is round, dendrite, axon disappeared, Nissl's staining, cel edema, deviation;Above the total dose of 80Gy,it showed more apoptotic cel s, large part of neurons lose polarity appearance, Nissl bodies disintegrated and disappeared, nuclear pyknosis, fragmentation, dissolution, apoptotic bodies. The Beagle spinal TUNEL staining showed: the total dose in the 80Gy fol owing a smal amount of apoptosis of nerve cel s, mainly in the anterior horn of spinal cord and white mat er; total dose in 80Gy above the visible visible apoptotic nerve cel s, mainly in the anterior horn of spinal cord and white mat er.Conclusion the normal Beagle spinal IGRT tolerance dose limit of about 80Gy.
