微生物法测定婴幼儿奶粉中游离生物素含量因实验室条件差异,严格按照国标方法测定标准曲线线性及结果的准确性及重现性均不太理想。对菌悬液浓度、菌种传代次数以及标准溶液配制方式进行优化,试验结果表明,最佳条件为菌悬液吸光度(A)0.50~0.64、菌种传代不超过5次、标准溶液使用超纯水临用现配的条件下标准曲线符合试验要求,相关系数(R2)可达0.9956,有较大的提高,此条件下平均回收率92%,变异系数为3.56%。

Because of the differences among the labs ,it could not satisfy the demands of accuracy and repeatability with microbiological method in national standard detecting the assay for biotin in infant formula milk powder.Microbial group concentration ,serial passages and the method of preparing standard solution were optimized. The results showed that the best condition was the absorbance of microbial group between 0.50 to 0.64 , serial passages less than five and using ultra pure water preparing standard solution ,the R2 could reach 0.995 6,the average recovery was 92%and the coefficient of variation was 3.56%.
