目的探讨microRNA-144在结肠癌中的表达水平及其与临床病理特征的相关性。方法提取38例结肠癌及其正常结肠黏膜组织标本的总 RNA,采用逆转录实时荧光定量聚合酶链反应(RT-qPCR)方法检测 microRNA-144的表达量,并分析其与临床分期、分化程度、淋巴结转移等临床病理特征的关系。结果癌与癌旁正常结肠黏膜组织比较,microRNA-144在癌组织中高表达,相对表达量的中位数是2.30。其表达与TNM分期(P<0.05)、分化程度(P<0.05)相关,受试者工作特征(ROC)曲线分析显示,以相对表达量1.70为临界点时,microRNA-144作为评价结肠癌细胞分化程度,敏感性为88%,特异性为61.5%。结论 microRNA-144在结肠癌中的异常表达可能与结肠癌的发生、发展有关。

목적탐토microRNA-144재결장암중적표체수평급기여림상병리특정적상관성。방법제취38례결장암급기정상결장점막조직표본적총 RNA,채용역전록실시형광정량취합매련반응(RT-qPCR)방법검측 microRNA-144적표체량,병분석기여림상분기、분화정도、림파결전이등림상병리특정적관계。결과암여암방정상결장점막조직비교,microRNA-144재암조직중고표체,상대표체량적중위수시2.30。기표체여TNM분기(P<0.05)、분화정도(P<0.05)상관,수시자공작특정(ROC)곡선분석현시,이상대표체량1.70위림계점시,microRNA-144작위평개결장암세포분화정도,민감성위88%,특이성위61.5%。결론 microRNA-144재결장암중적이상표체가능여결장암적발생、발전유관。
Objective To investigate the expression of microRNA-144 and its association with clinic-pathological features in co-lon tumor.Methods The expression levels of microRNA-144 in 38 colon tumors and their matched non-tumor adjacent tissue speci-mens were detected by using stem-loop real-time reverse transcription-polymerase chain reaction(RT-qPCR),and their correlation with clinic-pathologic features of colon tumor was analyzed.Results Expression levels of microRNA-144 were significantly higher in tumor tissues than in adjacent normal tissues(P<0.05),the median of comparative expression level was 2.30.The expression of microRNA-144 was associated with TNM stage(P<0.05)and cell differentiation(P<0.05).ROC curve of microRNA-144 about cell differentiation and resulted in a diagnostic sensitivity of 88% and specificity of 61.3%.Conclusion The level of microRNA-144 expression may be a valuable adj uvant parameter in forecasting the poorly differentiated colon tumor.
