目的:观察七氟烷预处理对急性缺氧小鼠脑氧化应激的影响,并探讨其与预处理的时间、剂量的关系。方法:60只雄性C57BL/6J小鼠随机分为6组:对照组(Control)、急性缺氧组(H组,6.5%O2,24 h)、2%Sevo 30 min+H组、2% Sevo 60 min+H组、4% Sevo 30 min+H组、Propofol+H组。按试剂盒说明检测脑匀浆丙二醛(MDA)含量及超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GPx)活性。结果:与H组比较,各七氟烷预处理组脑内MDA水平下降,并在2%Sevo 60 min+H组、4%Sevo 30 min+H组降低显著,Propofol+H组则无意义;GPx活性在各七氟烷预处理组有所升高,在4% Sevo 30 min+H组上升显著,Propofol+H组GPx活性下降,但不明显;与预计相反,各预处理使SOD活性显著降低。结论:七氟烷预处理减轻脑氧化应激而具有脑保护作用,并与时间、剂量相关。

목적:관찰칠불완예처리대급성결양소서뇌양화응격적영향,병탐토기여예처리적시간、제량적관계。방법:60지웅성C57BL/6J소서수궤분위6조:대조조(Control)、급성결양조(H조,6.5%O2,24 h)、2%Sevo 30 min+H조、2% Sevo 60 min+H조、4% Sevo 30 min+H조、Propofol+H조。안시제합설명검측뇌균장병이철(MDA)함량급초양화물기화매(SOD)、곡광감태과양화물매(GPx)활성。결과:여H조비교,각칠불완예처리조뇌내MDA수평하강,병재2%Sevo 60 min+H조、4%Sevo 30 min+H조강저현저,Propofol+H조칙무의의;GPx활성재각칠불완예처리조유소승고,재4% Sevo 30 min+H조상승현저,Propofol+H조GPx활성하강,단불명현;여예계상반,각예처리사SOD활성현저강저。결론:칠불완예처리감경뇌양화응격이구유뇌보호작용,병여시간、제량상관。
Aim:To investigate the effect of Sevoflurane(Sevo)preconditioning on oxidative stress of brain in acute hypoxia mice,and the relationship with duration of adminstering drug and concentration. Methods:Sixty male C56BL/6J mice were randomly allocated to 6 groups:Control,Hypoxia(H,6.5%O2 for 24 h),2%Sevo 30 min+H,2%Sevo 60 min+H,4%Sevo 30 min+H,Propofol+H.According to the kit instruction steps,the brain was homogenized for the measurement of malondialdehyde(MDA), superoxide dismutase(SOD),glutathione peroxidase(GPx).Results:Compared with H group,the brain levels of MDA reduced in all Sevo precondition groups,moreover,in the 2%Sevo 60 min+H group and 4% Sevo 30 min+H group,significantly reduced,however Propofol+H group was not statistically signif-icant;the activity of GPx increased in three Sevo precondition groups,and in the 4% Sevo 30 min+H group,this increased significantly,activity of GPx reduced in the Propofol+H group,but has not signifi-cant;the activity of SOD decreased in all precondition groups which was not influenced as expected.Con-clusion:Sevo preconditioning protects the mouse brain against oxidative stress,and the protection is relat-ed to duration of adminstering drug and dose.
