目的本研究旨在调查山东地区汉族人群醛糖还原酶(AR)5’端(AC)n二核苷酸重复序列多态性,探讨AR5’端(AC)n重复序列多态性与2型糖尿病(DM)、2型糖尿病合并冠心病(CHD)之间的关系。方法随机选取新诊断2型糖尿病患者74例,2型糖尿病合并冠心病者68例,正常对照组116例。应用聚合酶链式反应(PCR)、琼脂糖凝胶电泳技术、聚丙烯酰胺凝胶电泳技术及DNA测序检测AR等位基因,比较各组等位基因分布频率。结果3组均发现7种等位基因(Z-6~Z+6),各组间等位基因分布频率差异无统计学意义。结论 AR基因5’端(AC)n二核苷酸重复序列多态性等位基因不是糖尿病、糖尿病合并冠心病发病的独立危险因素。

목적본연구지재조사산동지구한족인군철당환원매(AR)5’단(AC)n이핵감산중복서렬다태성,탐토AR5’단(AC)n중복서렬다태성여2형당뇨병(DM)、2형당뇨병합병관심병(CHD)지간적관계。방법수궤선취신진단2형당뇨병환자74례,2형당뇨병합병관심병자68례,정상대조조116례。응용취합매련식반응(PCR)、경지당응효전영기술、취병희선알응효전영기술급DNA측서검측AR등위기인,비교각조등위기인분포빈솔。결과3조균발현7충등위기인(Z-6~Z+6),각조간등위기인분포빈솔차이무통계학의의。결론 AR기인5’단(AC)n이핵감산중복서렬다태성등위기인불시당뇨병、당뇨병합병관심병발병적독립위험인소。
Objective To investigate AR 5'end (AC)n polymorphism in Han Nationality population in Shandong district,and discuss its relationship between type 2 diabetes(T2DM),type 2 diabetes with coronary heart disease(CHD). Methods 74 patients with newly diagnosed type 2 diabetes mellitus, type 2 diabetes mellitus with coronary heart disease in 68 cases, 116 cases of normal control group were randomly selected. Polymerase chain reaction (PCR), agarose gel electrophoresis, polyacrylamide gel electrophoresis and DNA sequencing were used to detect AR alleles, allele frequency distribution in each group were compared. Results Three groups were found seven alleles (Z-6~Z+6), there were no obvious difference of allele frequency distribution between groups. Conclusion AR 5'end (AC)n polymorphisms may not the independent risk factors for diabetes mellitus, and diabetes combined with coronary heart disease.
