目的探讨靶向GST-π、polβ的siRNA重组慢病毒在裸鼠体内对人食管鳞癌顺铂耐药株耐药的逆转作用。方法采用皮下注射细胞法建立裸鼠人食管癌细胞细胞EC9706及其耐药细胞EC9706/cDDP移植瘤模型,观察其成瘤性和体内耐药性。利用靶向GST-π、polβ的siRNA重组慢病毒感染具有 MDR表型的裸鼠移植瘤,以期达到封闭GST-π、polβ的转录和表达,检测移植瘤细胞对药物的敏感性变化。结果 cDDP 联合靶向GST-π的siRNA慢病毒GSTsi2处理组中观察到移植瘤体积与cDDP组及PBS对照组比较,体积显著减小,表明经cDDP联合GSTsi2处理后,可明显恢复移植瘤细胞对 cDDP的敏感性。而在cDDP联合靶向polβ的siRNA慢病毒polsi1处理组中移植瘤体积与cDDP及PBS对照组比较无显著差异,移植瘤细胞对cDDP的敏感性无明显改变。结论靶向GST-π的重组慢病毒可有效逆转食管癌细胞的耐药性,靶向polβ的重组慢病毒逆转肿瘤细胞耐药的效果不明显。

목적탐토파향GST-π、polβ적siRNA중조만병독재라서체내대인식관린암순박내약주내약적역전작용。방법채용피하주사세포법건립라서인식관암세포세포EC9706급기내약세포EC9706/cDDP이식류모형,관찰기성류성화체내내약성。이용파향GST-π、polβ적siRNA중조만병독감염구유 MDR표형적라서이식류,이기체도봉폐GST-π、polβ적전록화표체,검측이식류세포대약물적민감성변화。결과 cDDP 연합파향GST-π적siRNA만병독GSTsi2처리조중관찰도이식류체적여cDDP조급PBS대조조비교,체적현저감소,표명경cDDP연합GSTsi2처리후,가명현회복이식류세포대 cDDP적민감성。이재cDDP연합파향polβ적siRNA만병독polsi1처리조중이식류체적여cDDP급PBS대조조비교무현저차이,이식류세포대cDDP적민감성무명현개변。결론파향GST-π적중조만병독가유효역전식관암세포적내약성,파향polβ적중조만병독역전종류세포내약적효과불명현。
Objective To investigate the reversal of cisplatin resistance of esophageal cancer by Lentiviral vector of siRNA targeting to GST-π, polβ in vivo. Methods EC9706 and EC9706/cDDP cells were subcutaneously injected into athymic nude mice to set up the animal models of grafted tumor. Injecting cDDP plus GSTsi2(polsi1) to xenografts derived from drug resistant cells to detect xenografts cells changes in drug sensitivity. Results The tumor volume of the group of cDDP plus GSTsi2 was lower than that of groups of PBS and cDDP; while the tumor volume of the group of cDDP plus polsi1 had no significant difference with groups of PBS and cDDP. Conclusion Injecting cDDP plus GSTsi2 to xenografts derived from drug resistant cells could inhibit its growth; while injecting cDDP plus polsi1 to xenografts could not inhibit its growth.
