目的 观测内脂素(Visfatin)在非酒精性脂肪肝病大鼠肝组织中的表达情况,拟为非酒精性脂肪肝病的诊治开阔出新的思路.方法 将40只SD大鼠按随机数字表法分为对照组和造模组,每组20只.对照组予基础饲料喂养,并每周两次腹腔内注射生理盐水(0.2 ml/100 g体质量);造模组予高脂饲料喂养,并每周两次腹腔内注射40% CCl4色拉油溶液(0.2 ml/100 g体质量).分析各组在2、4、6、8周4个时间点血清ALT、AST、TG、TC的变化及肝组织Visfatin的表达情况.结果 随着造模时间延长,造模组大鼠血清ALT、AST、TG、TC逐渐升高(P<0.05).免疫组化及RT-PCR显示造模组大鼠各时期肝组织中内脂素水平随着造模时间的延长而增高,较同期对照组比较差异有统计学意义(P<0.05).结论 (1)高脂喂养并腹腔内注射CCl4可以成功构建SD大鼠非酒精性脂肪肝病模型;(2)内脂素在非酒精性脂肪肝病大鼠肝脏组织中的表达水平随着肝脏脂肪变性程度增强而增高,因此可作为诊治非酒精性脂肪肝病并观测其严重程度的新指标.

Objective To investigate the expression of visfatin in the livers of rats suffering from non-alcoholic fatty liver disease (NAFLD) and to expect the development of new diagnosis and treatment for NAFLD.Methods A total of 40 Sprague-Dawley (SD) rats was randomly divided into control and NAFLD model groups.For control group,the rats were given basic diet and subcutaneous injection of saline(0.2 ml/100 g weight)twice a week.However,rats in the model group were given high fat diet and were given subcutaneous injection of 40 % carbon tetrachloride (CCl4) salad oil (0.2 ml/100 g) twice a week.The levels of alanine aminotransferase (ALT),aspartate aminotransferase (AST),triglyeride (TG),total-cholesterol (TC) in serum together with the visfatin expression in hepatic tissue were analyzed at the 2nd,4th,6th,and 8th weeks after operation,respectively.Results The levels of ALT,AST,TG,and TC in model group were increased gradually as time going(P < 0.05).Immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) experiments showed the level of visfatin in model group was significantly higher than control group in each period,with a increasing trends with the modeling time (P < 0.05).Conclusions (1)After high-fat feeding and intraperitoneal injection of CCL4,a model of adult SD rats suffering from NAFLD was constructed.(2)In livers of rats suffering from NAFLD,the expression of visfatin was increased while the level of liver steatosis was enhanced.Visfatin would be a new target to observe the severity of NAFLD.
